摘  要:背景:肥胖发生率逐渐攀升,易引起多种继发性疾病,对此,学者们提出众多减肥干预策略,其中运动干预或/和低氧暴露对肥胖防治具有较好的效果。目的:探讨低氧运动对肥胖大鼠骨骼肌过氧化物酶体增殖物激活受体α(peroxisome proliferators-activated receptorα,PPARα)/过氧化物酶体增殖物激活受体γ共激活分子1α(peroxisome proliferator-activated receptor-γcoactivator-1α,PGC-1α)/脂素1表达的影响。方法:构建营养性肥胖大鼠模型,建模后随机分为常氧安静组和运动组、体积分数16.3%低氧安静组和运动组、13.3%低氧安静组和运动组,继续高脂饲养,运动组进行8周耐力运动,20 m/min,40 min/d,每周5 d。末次运动24 h后麻醉处死大鼠并采样,Western blot及免疫组化测定大鼠骨骼肌PGC-1α、PPARα以及脂素1蛋白表达水平。结果与结论:①7周高脂膳食诱导营养性肥胖大鼠模型建立;②与常氧安静组相比,常氧运动组、16.3%低氧运动组及13.3%低氧运动组大鼠骨骼肌PGC-1α蛋白表达水平均显著升高(P<0.05-0.01);与16.3%低氧安静组相比,16.3%低氧运动组大鼠骨骼肌PGC-1α蛋白表达水平显著升高(P<0.05);与13.3%低氧运动组相比,常氧运动组、16.3%低氧安静组、13.3%低氧安静组大鼠骨骼肌PGC-1α蛋白表达水平均显著降低(P<0.05-0.01);③与常氧安静组相比,常氧运动组、13.3%低氧安静组和运动组大鼠骨骼肌PPARα蛋白表达水平显著升高(P<0.05-0.01);与13.3%低氧运动组相比,16.3%低氧安静组和运动组大鼠骨骼肌PPARα蛋白表达水平显著降低(P<0.05);④与常氧安静组相比,其他5组大鼠骨骼肌脂素1蛋白表达水平均有所降低,其中13.3%低氧运动组降低最为显著(P<0.05);⑤提示低氧运动可引起肥胖大鼠骨骼肌PGC-1α、PPARα蛋白表达增加,下调脂素1蛋白表达,促进骨骼肌脂肪酸氧化代谢,从而有利于改善肥胖大鼠骨骼肌的能量代谢。BACKGROUND:The incidence of obesity is gradually increasing,which is prone to cause a variety of secondary diseases.Therefore,numerous weight-loss intervention strategies have been proposed,among which exercise intervention or/and hypoxic exposure have better effects on the prevention and treatment of obesity.OBJECTIVE:To explo re the effect of hypoxic exercise on the protein expression levels of peroxisome proliferato rs-activated receptor α(PPARα)/peroxisome proliferato r-activated receptor-y coactivator-1α(PGC-1)/Lipin1 in the skeletal muscle of rats with alimentary obesity.METHODS:Animal models of alimentary obesity were established in rats,and the rat models were then randomized into normoxic quiet group and exe rcise gro up,16.3% hypoxic quiet group and exercise group,and 13.3% hypoxic quiet group and exercise group.The rats were fed with high fat,and all exe rcise gro ups received an endurance exercise,20 m/min,40 min/d,5 days per week for 8 wee ks.The rats were killed under anesthesia and sampled at 24 hours after the last exercise.The protein expression levels of PGC-1α,P PARα,and Lipin1 in the skeletal muscle of rats were then determined by western blot and immunohistochemistry.RESULTS AND CONCLUSION:The rat model of alimentary obesity was induced by a 7-week high-fat diet.Compared with the normoxic quiet group,the expression of PGC-1α protein in the skeletal muscle of rats was significantly increased in the normoxic exercise group,16.3% hypoxic exercise group,and13.3% hypoxic exercise group(P <0.05-0.01).Compared with the 16.3% hypoxic quiet group,the expression of PGC-1α protein in the skeletal muscle of rats was significantly increased in the 16.3% hypoxic exercise group(P<0.05).Compared with the 13.3% hypoxic exercise group,the expression of PGC-1α protein in the skeletal muscle of rats was significantly reduced in the normoxic exercise group,16.3% hypoxic quiet group,and 13.3% hypoxic quiet group(P <0.05-0.01).Compared with the normoxic quiet group,the expression of PPARα protein in the sk
