全真一气汤通过ACTA2调节COPD大鼠肺动脉平滑肌细胞收缩功能

摘　　要：目的:探讨全真一气汤通过ACTA2调节COPD大鼠肺动脉平滑肌细胞收缩功能的机制。方法:清洁级SD大鼠18只,采用烟熏的方法建立COPD大鼠模型,随机分成中药组与模型组,模型组大鼠每日予0.9%生理盐水5mL灌胃,中药组大鼠每日以全真一气汤浓缩汤剂0.4mL/20g的剂量进行灌胃干预,连续灌服60d。干预结束后,实时荧光定量PCR检测大鼠肺组织ACTA2表达水平的变化,Western blot检测大鼠肺组织中ACTA2蛋白水平的变化。结果:中药组ACTA2mRNA表达丰度为模型组的0.718倍,中药组ACTA2蛋白表达水平较模型组下调。结论:全真一气汤可能通过下调ACTA2表达影响COPD大鼠肺动脉平滑肌细胞收缩功能,延缓COPD病情进展。Objective:To investigate the mechanism of Quanzheng Yiqi Decoction regulating the contractile function of pulmonary artery smooth muscle cells in COPD rats through ACTA2.Methods:A COPD rat model was established by smoking.Eighteen male SD rats of clean grade were randomly divided into two groups:the traditional Chinese medicine group and the control group.The rats in the control group were given 5 mL 0.9% sodium chloride solution by gavage every day,while the rats in the traditional Chinese medicine group were given 0.4 mL/20 g of Quanzhen Yiqi Decoction concentrated decoction daily for 60 days.After the intervention,the expression of Actin Alpha 2(ACTA2)in lung tissue was detected by real-time fluorescent quantitative PCR.Western blot was used to detect the changes of Actin Alpha2(ACTA2)protein level in lung tissue.Results:The mRNA expression of ACTA2 in the control group was higher than that in the traditional Chinese medicine group,and its expression abundance was 0.718 in the control group;The expression of ACTA2 protein in the control group was up-regulated compared with the Chinese medicine group.Conclusion:Quanzhen Yiqi Decoction may affect the contractile function of pulmonary artery smooth muscle cells by down regulating the expression of ACTA2,thus delaying the progression of COPD.
