摘  要:沙门菌血清型4,[5],12:i:-作为鼠伤寒沙门菌单相变种的一个代表,自从20世纪90年代中期开始,在人类和动物中的流行不断增加。为调查沙门菌血清型4,[5],12:i:-在河南地区猪中的流行情况,本研究于2015年9月~2018年12月,从河南不同地区送检的63个病例中,分别采集发病仔猪的肺脏、心血、肝脏等样品,进行沙门菌的分离培养。通过染色镜检、PCR法对分离菌株进行鉴定,采用凝集法和鞭毛相抗原诱导转化方法鉴定分离菌株的血清型,采用双重PCR对沙门菌血清型4,[5],12:i:-分离菌株的鉴定结果进一步验证。采用肉汤稀释法测定分离菌株的耐药性。沙门菌的分离鉴定结果显示,从送检的63个病例中,共分离到6株沙门菌,阳性率为9.52%(6/63),其中3株为沙门菌血清型4,[5],12:i:-,另外3株为猪霍乱沙门菌和猪霍乱沙门菌孔成道夫变种。药敏试验结果显示,所有沙门菌分离菌株对氨苄青霉素、磺胺甲基异恶唑、萘啶酸和氯霉素均耐药,且呈多重耐药表型,其中沙门菌血清型4,[5],12:i:-分离菌株对卡那霉素、庆大霉素、阿奇霉素、四环素均耐药,而猪霍乱沙门菌和猪霍乱沙门菌孔成道夫变种分离菌株的耐药性较低。本研究首次报道沙门菌血清型4,[5],12:i:-在我国猪群中的流行情况,且其耐药性较强,可能给临床用药带来困难,对公共卫生安全构成潜在威胁。Salmonella serotype 4,[5],12:i:-represents a monophasic variant of Salmonella typhimurium,which became increasingly prevalent in both human and animal cases since the mid-1990 s.In order to investigate the prevalence of Salmonella serotype 4,[5],12:i:-in swine clinical cases in Henan,China,lung,blood and liver samples of diseased piglets were collected from 63 cases in different areas of Henan province between September 2015 and December 2018 for isolation and culturing of Salmonella.The isolates were identified by staining microscopy and PCR.The serotypes of the isolates were classified by an agglutination method and a method for flagellum phase reversal by induction.Then,Salmonella serotype 4,[5],12:i:-isolates were confirmed by duplex PCR.The drug resistance of the isolates was determined by the broth dilution method.The results of isolation and identification of salmonella showed that a total of 6 Salmonella strains were isolated from 63 cases,with an positive rate of9.52%(6/63),of which three strains were identified as Salmonella serotype 4,[5],12:i:-,and the other three strains were identified as Salmonella cholera-suis or Salmonella cholerae-suis var.kunzendorf.Results of drug susceptibility test showed that all of Salmonella isolates are resistant to ampicillin,sulfamethoxazole,nalidixic acid and chloramphenicol,with a multiple drug resistance phenotype;Salmonella serotype 4,[5],12:i:-isolates are resistant to kanamycin,gentamycin,azithromycin and tetracycline;however,Salmonella cholera-suis isolates or Salmonella cholerae-suis var.kunzendorf isolates have much lower resistance rates to most of drugs.This study for the first time reports the prevalence of Salmonella serovar 4,[5],12:i:-in diseased swine in China,and drug resistance of these isolates is severe,which may bring difficulties in choosing clinical medication to treat pigs.Furthermore,these Salmonella serotype 4,[5],12:i:-isolates from diseased swine can pose a potential threat to public health security.
