蒙药植物耧斗菜总生物碱的提取与鉴定

摘　　要：提取蒙药用植物耧斗菜的总生物碱,并对其进行定性检测与结构鉴定。采用超声波辅助提取法提取耧斗菜总生物碱,通过4种沉淀试剂反应定性检测生物碱,用气相-质谱(GC-MS)联用技术进行结构鉴定。结果发现耧斗菜总生物碱提取液对4种生物碱定性试剂均有阳性反应,生物碱含量为0.155 mg/g;从耧斗菜提取物中鉴定出2种希夫碱类化合物,分别是酰腙类生物碱2,6-二甲氧基苯甲醛氨甲酰腙和酰肼类生物碱2-(3-对氯苯氧)N-(-2,4二甲氧基苄叉)乙酰肼。耧斗菜富含有生物碱类化合物,在生物碱提取开发利用方面有较好的应用前景。The columbine(Aquilegia viridiflora Pall),a kind of Mongolian medicinal plant,is rich in alkaloid,and is a suitable material for the extraction,development and utilization of alkaloid. In this study,the total alkaloids were extracted by ultrasonic assisted method from columbine,and the type of them was identified through precipitation reaction by four reagents,the structure of them was determined by gas chromatographymass spectrometry(GC-MS). The results showed that the content of the total alkaloid extracted from columbine was 0. 155 mg/g and the total alkaloid had positive reactions with four alkaloid precipitation reagents. Two Schiff bases were identified by GC-MS in the extracts of columbine,which were acylhydrazone alkaloid(2,6-dimethoxybenzylcarbazone) and acylhydrazine alkaloid[2-(3-p-chlorophenoxy) N-(-2,4-dimethoxybenzyl)acetyl hydrazine]. The research laid a basic foundation for the extraction of alkaloids from the Aquilegia viridiflora pall.
