摘 要:目的:基于网络药理学探究化湿败毒方治疗新型冠状病毒肺炎的作用靶点及分子作用机制。方法:从ETCM、TCMSP、BATMAN-TCM等平台获取化湿败毒方的药物性、味、归经及活性成分、匹配靶标并进行筛选。从GENE CARDS平台得到新型冠状病毒肺炎(COVID-19)靶点;从STRING平台获取靶标网络,并进行MCODE模块分析,筛选关键靶标;使用METASCAPE平台进行基因本体论(GO)分析与京都基因和基因组数据库(KEGG)通路分析。结果:共筛选到224个药物活性成分,178个对应靶标。得到COVID-19的潜在作用靶标26个,作用于739项生物过程和97条通路。结论:化湿败毒方可能通过干预细胞因子相关通路,达到抗炎、抑制细胞因子风暴、干预机体免疫等作用,以多个靶点、成分、途径,多方面治疗重型COVID-19。Objective:To explore the targets and molecular mechanism of Huashi Baidu Formula(化湿败毒方)in the treatment of COVID-19 based on network pharmacology.Methods:The medicinal property,flavor,channel tropism,active components and corresponding targets of Huashi Baidu Formula was screened by using the databases of ETCM,TCMSP,BATMAN-TCM.The COVID-19 targets were obtained from the GENE CARDS database.The target network was obtained from the STRING database,then conducted MCODE module analysis to select the key targets.Finally,the Metascape database was used to carry out Gene Ontology(GO)analysis and Kyoto Gene and Genomic Encyclopedia(KEGG)pathway analysis.Results:Total 224 active components and 178 corresponding targets were screened out.Total 26 potential targets of COVID-19 were obtained,acting on 739 biological processes and 97 pathways.Conclusions:Huashi Baidu Formula may achieve anti-inflammatory,inhibit cytokine storm and immune intervention by intervening cytokine related pathway,and treat severe type of COVID-19 with multiple targets,components and approaches.
