摘 要:目的探索干扰素刺激基因(STING)在氟氯氰菊酯致大鼠睾丸损伤中的作用。方法40只SPF级雄性Wistar大鼠随机分为4组,即对照组、氟氯氰菊酯低剂量组、中剂量组、高剂量组,每组10只。染毒4周后,HE染色观察大鼠睾丸组织形态变化;透射电镜观察睾丸细胞超微结构变化;采用试剂盒测定睾丸组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)及乳酸脱氢酶(LDH)含量的变化;采用Western blot、qPCR检测STING、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)蛋白及mRNA表达水平变化。结果HE染色结果显示:与对照组相比,随着氟氯氰菊酯浓度增加,睾丸组织生精小管结构破坏越严重,生精小管管腔内精子有不同程度的减少。透射电镜结果显示:与对照组相比,染毒组氟氯氰菊酯浓度越高,精子细胞内线粒体肿胀越严重。与对照组相比,中、高剂量组LDH含量均增加(F=12.87,P<0.05),低、中、高剂量组MDA含量均升高(F=9.465,P<0.05);中、高剂量组中SOD活力降低(F=12.42,P<0.05)。Western blot显示,与对照组相比,氟氯氰菊酯低、中、高剂量组中STING、IL-6、TNF-α的蛋白表达水平均上调(P均<0.05)。与对照组相比,氟氯氰菊酯低、中、高剂量组STING、IL-6、TNF-α的mRNA表达水平均升高(P均<0.05)。结论在氟氯氰菊酯诱导大鼠睾丸损伤中,STING可能正向调控炎症因子TNF-α与IL-6的表达促进睾丸炎性反应。Objective To explore the role of STING gene in cyfluthrin-induced testicular injury in rats.Methods Forty SPF male wistar rats were randomly divided into 4 groups,namely,control group,low-dose group,medium-dose group,and high-dose group,with 10 rats in each group.After 4 weeks of exposure,HE staining was used to observe the morphological changes of rat testis;transmission electron microscopy was used to observe the ultrastructural changes of testis cells;the test kit was used to determine superoxide dismutase(SOD),malondialdehyde(MDA)and changes in lactate dehydrogenase(LDH)content;Western blot and qPCR were used to detect protein and mRNA expression levels of STING,IL-6,TNF-α.Results The results of HE staining showed that compared with the control group,as the concentration of the poison increases,the structure of the seminiferous tubules in the testicular tissue was damaged more severely,and the sperm in the seminiferous tubules decreased to varying degrees.The results of transmission electron microscopy showed that compared with the control group,the higher the concentration of cyfluthrin in the infected group,the more severe the swelling of mitochondria in sperm cells.Compared with the control group,the LDH content in the medium and high-dose exposure groups increased(F=12.87,P<0.05),and the MDA content in the low,medium,and high-dose exposure groups increased(F=9.465,P<0.05);SOD activity decreased in the medium and high-dose exposure groups(F=12.42,P<0.05).Western blot showed that compared with the control group,the expression levels of STING,IL-6,TNF-αin the low,medium and high-dose of cyfluthrin were up-regulated(P<0.05).Compared with the control group,the expression levels of STING,IL-6,TNF-αmRNA in the low,medium and high-dose groups of cyfluthrin increased(P all<0.05).Conclusion In cyfluthrin-induced testicular injury of rats,STING may positively regulate the expression of inflammatory factors TNF-αand IL-6,and promote testicular inflammation.
