摘 要:目的研究低水平林丹外源性暴露对雌性SD大鼠卵巢及子宫功能形态的影响。方法选取健康6~8周龄的雌性SD大鼠45只,随机分为5组,每组9只。参照水系沉积物中林丹残留浓度2μg·g-1(除海河、辽河等高残留水系),将实验组分别设为林丹低(3 mg·kg^(-1))、中(6 mg·kg^(-1))、高剂量组(13 mg·kg^(-1))、阳性对照组(100μg·kg^(-1)雌二醇)以及溶剂对照组(1 mL玉米油)。腹腔注射10%水合氯醛麻醉后解剖取材,计算子宫、卵巢器官系数,测定促性腺激素释放激素(GnRH)、卵泡刺激素(FSH)、孕酮(P)的分泌水平,观察子宫、卵巢的组织形态学变化。结果林丹各剂量组大鼠体质量增长量与阳性对照组相比差异均无统计学意义(P均>0.05);低剂量实验组大鼠与溶剂对照组相比差异无统计学意义(P>0.05);中、高剂量实验组体质量增长量均低于溶剂对照组(P均<0.05)。各实验组大鼠子宫、卵巢质量及子宫、卵巢系数与阳性对照组、溶剂对照组相比差异均无统计学意义(P均>0.05)。林丹各剂量组血清GnRH、FSH、P激素分泌水平均低于溶剂对照组(P均<0.05);林丹各剂量组血清GnRH、FSH、P激素分泌水平与阳性对照组相比差异均无统计学意义(P均>0.05)。各实验组大鼠卵泡结构受损,闭锁卵泡明显增多,颗粒细胞数量减少,排列稀疏,胞核大小不一且伴有不同程度损伤。各实验组大鼠子宫内膜出现不同程度的胞核假复层现象,上皮组织增生,上皮细胞排列不均,胞核大小不一,并且随染毒剂量升高,子宫上皮出现腺体数量减少。结论低水平林丹可诱导颗粒细胞凋亡,使血清性激素GnRH、FSH、P分泌水平下降,子宫、卵巢等生殖器官发生组织形态及功能改变。Objective This experiment investigated the effects of low dose lindane on ovarian and uterine function and morphology.Methods 45 healthy female SD rats aged 6-8 weeks were randomly divided into 5 groups(n=9):solvent control group(1 mL corn oil),positive control group(100μg·kg^(-1)estradiol),and 3,6,and 13 mg·kg^(-1)lindane groups.Refer to the residual concentration of lindane in stream sediments 2μg·g-1(except high residual water systems such as Haihe River and Liaohe River).After an overnight fasting,10%chloral hydrate was injected intraperitoneally,and the rats were dissected after anesthesia.The levels of serum sex hormones and organ coefficient of uterus and ovary were determined.Determined Organ coefficient of uterus and ovary and the secretion levels of gonadotropin releasing hormone,follicle stimulating hormone and progesteron.The histomorphological changes of uterus and ovary were observed.Results Compared with the positive control group,there was no significant difference in body weight of rats in the exposure groups(P all>0.05);Compared with the solvent control group,there was no significant difference in body weight of rats in the 3 mg·kg^(-1)groups(P>0.05);Compared with the solvent control group,the body weight of rats in the 6,13 mg·kg^(-1)groups decreased(P all<0.05).Compared with the positive control group and Solvent control group,there was no significant difference in the quality of uterus and ovary and the coefficient of uterus and ovary in the exposure groups(P all>0.05).Compared with the solvent control group,the GnRH,FSH and P hormone concentration decreased in exposure groups(P all<0.05);compared with the positive control group,there was no significant difference in body weight of rats in the exposure groups(P all>0.05).There was no significant difference in the GnRH,FSH and P hormone concentration of rats in the exposure groups(P all>0.05).The follicular structure of rats was damaged,the number of atretic follicles increased significantly,the number of granulosa cells decreased and a
