摘　　要：目的:探究橄榄苦苷对肝脏缺血再灌注损伤(HIRI)的保护作用。方法:采用肝蒂夹紧60min再灌注90min的方式制作大鼠HIRI模型,随机分成假手术组(A组)、对照组(B组)、围术期橄榄苦苷治疗组(C组)、术前橄榄苦苷治疗组(D组),C组和D组分别在术前2h和术前10d予橄榄苦苷40mg·kg-1·d-1。收集各组大鼠血液和肝组织样本,进行肝功能指标、氧化应激水平、炎症因子和免疫指标比较以及组织病理学的评估。结果:C组和D组的肝功能、组织氧化应激参数、炎症因子和免疫指标均显著低于B组(P<0.05);此外,C组和D组中组织病理学损伤显著降低。结论:橄榄苦苷能够减轻HIRI的氧化应激和炎症反应,提高免疫功能,具有明显的肝脏保护作用。Objective:To explore the protective effect of oleuropein on hepatic ischemia-reperfusion injury(HIRI).Methods:The rat HIRI model was made by clamping the liver pedicle for 60 minutes and reperfusion for 90 minutes.All the rats were randomly divided into 4 groups including the sham operation group(group A),control group(group B),perioperative oleuropein treatment group(group C),preoperative oleuropein treatment group(group D),in which rats in group C and D were given 40 mg·kg-1·d-1 oleuropein 2 hours before operation and 10 days before operation respectively.Blood and liver tissue samples of rats were collected to compare changes in liver function,oxidative stress level,inflammatory factors,immune indicators and histopathology in the four groups.Results:The liver function,parameters of tissue oxidative stress,inflammatory factors and immune indicators of the group C and D were significantly lower than those of group B(P<0.05).And the histopathological damages of the group C and D were also significantly reduced.Conclusion:Oleuropein showed obvious liver-protective effect through alleviating oxidative stress response and anti-inflammatory response,improving immune function.
