摘 要:本文通过聚乙二醇化合成了酶-高分子共聚物纳米颗粒(mPEG-GO;&HRP conjugate nanoparticles,mCNPs),并将其固载在纸芯片上,制备了一种便携式的葡萄糖比色纸芯片。通过扫描电镜(SEM)和能谱分析(EDS)对mCNPs纸芯片的形貌和元素进行了表征和分析。结果表明,mCNPs已成功负载在纸纤维上。在优化的检测条件下,微克级的mCNPs被固载在纸芯片上,可快速(30 s左右)实现0.1~3.0 mmol/L浓度范围内的葡萄糖的比色检测,检出限为0.01 mmol/L。在高温环境和长期储存时,与游离酶纸芯片相比,mCNPs纸芯片表现出更稳定的检测性能。本方法用于检测真实血清样本的回收率为97.52%~101.76%,具备潜在的实际应用价值。In this work,enzyme-polymer conjugate nanoparticles(mPEG-GO;&HRP conjugate nanoparticles,mCNPs)were fabricated using PEGylation,which were anchored on the paper to prepare a portable glucose colorimetric microchip.The morphology and elemental composition of mCNPs paper were characterized by SEM and EDS,and the output indicated that mCNPs were successfully loaded on the paper fiber.The results show that,in the optimized detection conditions,even several micrograms of mCNPs mounted on a paper microchip can achieve colorimetric detection of glucose with the concentration range from 0.1 mol/L to 3.0 mmol/L in a short time(about 30 s),and the limit of detection is 0.01 mmol/L.Further experimental data show that,in the condition of high-temperature or long-term storage,mCNPs paper exhibits better stability,compared with those paper microchips based on free enzyme.The high recovery rate of this method in detection of real serum samples(97.52%-101.76%)also enables it to be a good glucose colorimetric paper in the future.
