摘 要:H_(2)O_(2)与NO是在多种生理条件下产生的强氧化剂和硝化剂,二者的平衡与过量产生与多种神经退行性疾病的发生有关。因此,同时监测生物系统中H_(2)O_(2)与NO的浓度对了解二者在生理条件下的作用机制具有重要意义。基于电化学方法快速、准确、灵敏度高等优点,本研究提出了一种可在体同步检测H_(2)O_(2)和NO的双微电极。将铂纳米颗粒(PtNPs)和邻苯二胺(OPD)用于修饰碳纤维(CF)电极,以提高电极的灵敏度和选择性。同时将参比电极与工作电极集成在一起,减小电极体积,减轻电极对组织细胞的损伤。测得H_(2)O_(2)电极的线性检测范围为2.0~96.9μmol/L(R^(2)=0.9904),检测限为1.7μmol/L;NO电极的线性检测范围为2.1~279.3μmol/L(R^(2)=0.9983),检测限为0.5μmol/L。该电极对抗坏血酸、多巴胺等体内常见干扰物没有响应,具有较强的抗干扰能力。H_(2)O_(2) and NO are strong oxidants and nitrifiers produced under many physiological conditions.The balance and overproduction of H_(2)O_(2) and NO are related to the occurrence of a variety of neurodegenerative diseases.Therefore,monitoring the concentrations of H_(2)O_(2) and NO in biological systems at the same time is important to know the mechanism of action under physiological conditions.Based on the advantages of electrochemical methods such as fast,accurate,and high sensitivity,this study proposes a dual microelectrode that can simultaneously detect H_(2)O_(2) and NO in vivo.The surface of the carbon fiber electrode is modified with platinum particles and o-phenylenediamine to improve the sensitivity and selectivity of the electrode.At the same time,the reference electrode and the working electrode are integrated to reduce the damage of the electrode to the tissue cells.The linear detection range of the H_(2)O_(2) electrode is 2.0-96.9μmol/L(R^(2)=0.9904),and the detection limit is 1.7μmol/L.The linear detection range of the NO electrode is 2.1-279.3μmol/L(R^(2)=0.9983),the detection limit is 0.5μmol/L.The electrode does not respond to common interferences in the body such as ascorbic acid and dopamine,and has strong anti-interference ability.
