针刺对肥胖大鼠脂肪组织M1型巨噬细胞及IL-6表达的影响研究

摘　　要：目的:观察针刺对肥胖大鼠脂肪组织M1型巨噬细胞及IL-6表达的影响,探讨针刺治疗肥胖的分子作用机制。方法:将24只SD雄性大鼠随机分为正常组、模型组和针刺组,每组8只。予“双侧足三里、阴陵泉、丰隆、天枢、中脘及关元”等穴位针刺,30 min/次/d,每5 d暂停1次,共30 d,并记录每周体质量。治疗结束后,分离各组大鼠的肾周脂肪、肠周脂肪及附睾脂肪等内脏脂肪,并记录其湿重。采用蛋白质免疫印迹(Western Blot)技术检测各组大鼠脂肪组织中M1型巨噬细胞标志物诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)及IL-6蛋白的表达;HE染色观察各组大鼠附睾脂肪组织形态。结果:与正常组比较,模型组大鼠的体质量、内脏脂肪湿重、iNOS及IL-6蛋白的表达均显著升高,脂肪细胞平均面积增大,差异具有统计学意义(P<0.05或P<0.01)。与模型组比较,针刺组大鼠的体质量、内脏脂肪湿重、iNOS及IL-6蛋白的表达均显著降低,脂肪细胞平均面积减小,差异具有统计学意义(P<0.05或P<0.01)。结论:针刺能降低肥胖大鼠脂肪组织M1型巨噬细胞标志物iNOS及IL-6的表达,改善机体的慢性炎症状态,以达到降脂减肥的目的。Objective:To observe the effect of acupuncture on the protein expressions of M1-type macrophages of iNOS and IL-6 in fat tissues of obese rats,and to explore the molecular mechanism of acupuncture in treatment of chronic obesity.Methods:24 SD male rats were randomly divided into the normal group,the model group and the acupuncture group,with 8 rats in each group.Acupuncture was performed at the bilateral acupoints of Zusanli(ST36),Yinlingquan(SP9),Fenglong(ST40),Tianshu(ST25),Zhongwan(RN12)and Guanyuan(RN4)for 30 minutes.The body mass was recorded every week.After the treatment,visceral fat,such as perirenal fat,periintestinal fat and epididymal fat,was isolated in each group and their wet weight was recorded.Western Blot was used to detect the protein expressions of M1-type macrophages of iNOS and IL-6 in adipose tissues of rats in each group.The morphology of epididymal adipose tissues was observed by HE staining.Results:Compared to those in the normal group,the body mass,the visceral fat wet weight,the protein expressions of iNOS and IL-6 were significantly increased,and the adipocyte diameter was also increased in the model group(P<0.05,P<0.01).Compared to those in the model group,the body mass,the visceral fat wet weight,the protein expressions of iNOS and IL-6 were significantly decreased,and the adipocyte diameter was also decreased in the acupuncture group(P<0.05,P<0.01).Conclusion:Acupuncture can reduce the expressions of M1-type macrophages of iNOS and IL-6,improve the chronic inflammatory state,and achieve the purpose of reducing fat and weight in obese rats.
