摘 要:目的研究纳达合剂治疗胆汁反流性胃炎的作用机制。方法将60只SD大鼠随机分为对照组、模型组、纳达低剂量组、纳达中剂量组、纳达高剂量组和麦滋林组,每组10只。除对照组外,其余组大鼠均采用灌胃反流液(牛胆酸钠+胰酶+卵磷脂)法制备胆汁反流性胃炎模型。造模4周后,纳达低、中、高剂量组在反流液灌胃的基础上给予12.75 g生药/kg、25.5 g生药/kg、38.25 g生药/kg纳达合剂灌胃,麦滋林组在反流液灌胃的基础上给予0.33 g/kg麦滋林药液灌胃,均1次/d,连续灌胃4周。MitoSOX Red探针检测大鼠胃黏膜线粒体活性氧(ROS)生成情况,Real-time PCR法及Western blot法检测大鼠胃黏膜组织中NLRP3、凋亡相关斑点样蛋白(ASC)、炎性半胱天冬酶-1(Caspase-1)mRNA和蛋白表达情况,ELISA法检测大鼠胃黏膜组织中白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)含量。结果模型组大鼠胃黏膜线粒体ROS含量,胃黏膜组织中NLRP3、ASC、Caspase-1 mRNA和蛋白表达量,胃黏膜组织中IL-1β、IL-18含量均显著高于对照组(P均<0.05);麦滋林组和纳达低、中、高剂量组上述各指标均显著低于模型组(P均<0.05),且纳达各组各指标呈剂量依赖性降低,两两比较差异均有统计学意义(P均<0.05)。结论纳达合剂对胆汁反流性胃炎大鼠的治疗作用可能与抑制ROS-NLRP3通路有关。Objective It is to study the mechanism of Nada mixture in the treatment of bile reflux gastritis.Methods Sixty SD rats were randomly divided into control group,model group,Nada mixture low-dose group(12.75 g crude drug/kg),Nada mixture medium-dose group,Nada mixture high-dose group and Maizilin group,10 rats in each group.Except for the control group,the rats in the other groups were given reflux liquid of sodium cholate,trypsin and lecithin by gavage to establish models of bile reflux gastritis.After 4 weeks of model establishment,the Nada low,medium and high dose groups were respectively given 12.75 g crude drug/kg,25.5 g crude drug/kg,38.25 g crude drug/kg Nada mixture by gavage on the basis of reflux fluid,while the Methrin group was given 0.33 g/kg Mezulin by gavage on the basis of reflux fluid,all once a day,continuously treated for 4 weeks.MitoSOX Red probe was used to detect the generation of reactive oxygen species(ROS)in the gastric mucosa of rats,and Real-time PCR and Western blot methods were used to detect NLRP3,apoptosis-associated speck-like protein(ASC),inflammatory cysteine(Caspase-1)mRNA and protein expression in gastric mucosa of rats,ELISA method was used to detect the content of interleukin-1β(IL-1β)and interleukin-18(IL-18)in gastric mucosa of rats.Results The content of mitochondrial ROS in gastric mucosa,the mRNA and protein expressions of NLRP3,ASC,Caspase-1 in gastric mucosa,and the contents of IL-1βand IL-18 in gastric mucosa of rats in the model group were significantly higher than those in the control group(all P<0.05),the above indexes in the Mezulin group and the Nada low,medium and high dose groups were significantly lower than those in the model group(all P<0.05),and the indexes in each Nada group decreased in a dose-dependent manner,the differences were significant(all P<0.05).Conclusion The therapeutic effect of Nada mixture on bile reflux gastritis in rats may be related to the inhibition of ROS-NLRP3 pathway.
