摘  要:目的:探讨白杨素对7-酮基胆固醇(7-KC)所致RAW264.7细胞炎症和脂代谢紊乱过程中的作用及可能机制。方法:采用MTT法检测细胞活性,筛选白杨素的安全给药浓度。以2μg/mL 7-KC孵育RAW264.7细胞从而构建巨噬细胞脂代谢紊乱模型,并设置空白组、模型组、瑞舒伐他汀组以及白杨素高(5μmol/L)、中(1μmol/L)、低(0.1μmol/L)剂量组。采用硝酸盐还原酶法检测各组细胞上清液中一氧化氮(NO)分泌量;实时荧光定量PCR(RT-qPCR)法检测ABCG1、LXRα、IL-17A、IL-10、IL-6、MCP-1的mRNA表达水平;western blotting法检测NF-κB P65、iNOS、SR-B1的蛋白表达水平。结果:与模型组相比,白杨素组细胞分泌的NO减少(均P<0.05),细胞内IL-6、MCP-1的mRNA和NF-κB P65、iNOS的蛋白表达水平降低,细胞内ABCG1、LXRα、IL-17A、IL-10的mRNA和SR-B1的蛋白表达水平升高(均P<0.05)。结论:白杨素可抑制7-KC诱导的巨噬细胞炎症反应,调节脂质转运相关蛋白和基因,其作用机制可能与抑制NF-κB的激活有关。Objective:To investigate the effect of chrysin on inflammation and lipid metabolism disorder of RAW264.7 cells induced by 7-keto-cholesterol(7-KC).Metheds:To choose an appropriate concentration for subsequent experimental treatments,we examined the viability of RAW264.7 cells by MTT assay.The RAW264.7 cells were cultured with 2μg/mL 7-KC to induce lipid metabolism disorder.Cells were divided into blank group,model group,rosuvastatin group,and different doses of chrysin(5μmol/L,1μmol/L,0.1μmol/L)groups.The nitric oxide(NO)content in supernatant was determined by nitrate reductase method.The mRNA expression levels of ABCG1,LXRα,IL-17A,IL-10,IL-6 and MCP-1 were detected by RT-qPCR.The protein expression levels of NF-κB P65,iNOS and SR-B1 were detected by western blotting.Results:Compared with the model group,the NO content,the mRNA expressions of IL-6 and MCP-1 and the protein expressions of NF-κB P65 and iNOS were decreased,while the mRNA expressions of ABCG1,LXRα,IL-17A and IL-10 and the protein expression of SR-B1 were increased(P<0.05).Conclusion:Chrysin can inhibit 7-KC induced macrophage inflammation and regulate lipid transport-related genes and proteins expressions.The mechanism may be related to the inhibition of NF-κB activation.
