摘 要:目的:观察原花青素对烫伤大鼠创面的治疗效果及其对血管内皮生长因子(VEGF)、血管内皮生长因子受体2(VEGFR2)的影响。方法:建立烫伤大鼠模型,将120只SD雄性大鼠随机分为健康组、烫伤组、原花青素高剂量组、原花青素低剂量组、阳性对照组、原花青素+VEGF抑制组,每组20只。记录各组大鼠1 d、5 d、10 d、15 d、20 d创面愈合情况;统计各组上皮出现时间/完全愈合时间;选取15 d的创面组织行苏木精—伊红(HE)染色,观察皮肤组织病理学变化情况;采用荧光定量PCR及western blotting法检测各组大鼠愈合创面组织中VEGF、VEGFR2 mRNA和蛋白表达水平。结果:健康组具有完整的皮肤组织,烫伤组皮肤创面组织坏死严重,烫伤组大鼠创面组织VEGF、VEGFR2 mRNA和蛋白表达显著高于健康组(P<0.05);原花青素高剂量组、原花青素低剂量组、阳性对照组大鼠创面逐渐恢复,创面愈合率显著高于烫伤组,上皮出现时间、完全愈合时间显著缩短,VEGF、VEGFR2 mRNA和蛋白表达显著升高(均P<0.05);原花青素高剂量组大鼠创面愈合程度最高,创面愈合率显著高于原花青素低剂量组、阳性对照组、原花青素+VEGF抑制组,上皮出现时间、完全愈合时间显著缩短,VEGF、VEGFR2蛋白表达显著高于原花青素低剂量组、原花青素+VEGF抑制组(均P<0.05),与阳性对照组相比无显著差异(P>0.05)。结论:原花青素可有效促进VEGF、VEGFR2 mRNA和蛋白表达,加速创面组织的恢复,从而缓解烫伤危害。Objective:To observe the therapeutic effects of procyanidin on the wounds of scalded rats and its effects on vascular endothelial growth factor(VEGF)and vascular endothelial growth factor receptor 2(VEGFR2).Methods:Scalded rat models were established.120 SD male rats were randomly divided into healthy group,scalded group,high-dose procyanidin group,low-dose procyanidin group,positive control group,and procyanidin+VEGF inhibitor group,with 20 rats in each group.The wound healing of rats in each group was recorded at the 1st,5th,10th,15th,and 20th day;The appearance time/complete healing time of epithelium in each group werecounted.The wound tissue was selected at the 15th day for hematoxylin-eosin(HE)staining to observe the pathological changes of skin tissue.Fluorescence quantitative PCR and western blotting were used to detect the expression levels of VEGF and VEGFR2 mRNA and protein in the wound tissues.Results:The healthy group rats had complete skin tissues,the wound tissues of the scald group had severe necrosis,and the expressions of VEGF and VEGFR2 mRNA and protein in the wound tissues of the scald group were significantly higher than those of the healthy group(P<0.05).The wounds of rats in the high-dose procyanidin group,low-dose procyanidin group,and positive control group gradually recovered,the wound healing rate was significantly higher than that of the scald group,the appearance time and complete healing time of epithelium were significantly shortened,and the expressions of VEGF and VEGFR2 mRNA and protein weresignificantly increased(P<0.05).The high-dose procyanidin group had the highest degree of wound healing,and the wound healing rate was significantly higher than that of the low-dose procyanidin group,positive control group,and procyanidin+VEGFinhibitorgroup.The appearance time and complete healing time of epithelium were significantly shortened,the expressions of VEGF and VEGFR2 proteins werehigher than that of the low-dose procyanidin group and procyanidin+VEGFinhibitorgroup(P<0.05),and there
