摘  要:目的:探讨颗粒蛋白前体(PGRN)在植入性胎盘组织中的表达以及PGRN对人绒毛膜滋养层细胞HTR-8/SVneo迁移、侵袭的影响及潜在的作用机制。方法:收集河北省中医院收治的植入性胎盘组织和正常胎盘组织各25例,实时荧光定量PCR(qRT-PCR)和蛋白免疫印迹(Western Blot)检测胎盘组织(绒毛膜层)中PGRN mRNA和蛋白水平;体外培养人绒毛膜滋养层细胞HTR-8/SVneo,采用重组腺病毒载体和小干扰RNA(siRNA)技术过表达和干扰HTR-8/SVneo细胞中的PGRN,通过qRT-PCR和Western Blot检测细胞中PGRN表达确定过表达和干扰有效后,划痕实验和Transwell实验分别检测HTR-8/SVneo细胞迁移、侵袭能力;Western Blot检测HTR-8/SVneo细胞迁移、侵袭相关蛋白的表达。结果:植入性胎盘组织中PGRN mRNA和蛋白表达水平均显著高于正常胎盘组织(P<0.05);过表达PGRN后,HTR-8/SVneo细胞迁移、侵袭能力显著增加(P<0.05),N-钙黏蛋白(N-cadherin)、纤维连接蛋白(FN)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)蛋白表达水平显著升高,E-钙粘蛋白(E-cadherin)表达水平显著降低(P<0.05),而PGRN沉默后,HTR-8/SVneo细胞的迁移、侵袭能力显著降低(P<0.05),同时N-cadherin、FN、MMP-2、MMP-9蛋白表达水平显著降低,E-cadherin蛋白表达水平显著升高(P<0.05)。结论:PGRN在植入性胎盘组织中过表达,PGRN的过表达可能促进人绒毛膜滋养层细胞HTR-8/SVneo的迁移和侵袭;而下调PGRN的表达能抑制人绒毛膜滋养层细胞HTR-8/SVneo的迁移和侵袭;其作用机制可能与促进上皮-间质转化(EMT)过程有关。Objective:To investigate the expression of progranulin(PGRN)in placenta accreta tissue and the effect of PGRN on the migration and invasion of human villous trophoblasts HTR-8/SVneo cells and its potential mechanism.Methods:Twenty-five cases of placenta accreta tissue and normal placenta tissue were collected in Hebei Hospital of Traditional Chinese medicine, the levels of PGRN mRNA and protein in placenta tissue(chorion layer) were detected by real time fluorescent quantitative PCR(qRT-PCR) and Western Blot, respectively;human villous trophoblasts HTR-8/SVneo was cultured in vitro, recombinant adenovirus vector and small interfering RNA(siRNA) technology were used to overexpress and interfere PGRN in HTR-8/SVneo cells, respectively.The effect of overexpression and interference of PGRN in cells were confirmed by qRT-PCR and Western blot, the migration and invasion ability of HTR-8/SVneo cells were detected by scratch test and Transwell test, respectively;Western blot was used to detect the expression of migration and invasion related proteins in HTR-8/SVneo cells.Results:The levels of PGRN mRNA and protein in placenta accreta tissue were significantly higher than those in normal placenta tissue(P<0.05);after overexpression of PGRN,the migration and invasion of HTR-8/SVneo cells were significantly increased(P<0.05),the expression levels of N-cadherin, fibronectin(FN),matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)were significantly increased, and the expression level of E-cadherin was significantly decreased(P<0.05).After PGRN silencing, the migration and invasion of HTR-8/SVneo cells were significantly decreased(P<0.05),at the same time, the expression levels of N-cadherin, FN, MMP-2 and MMP-9 were significantly decreased, and the expression level of E-cadherin was significantly increased(P<0.05).Conclusions:The level of PGRN in placenta accreta tissue is higher than normal placenta tissue.Overexpression of PGRN may promote the migration and invasion of HTR-8/SVneo cells;in contrast, down-r
