摘  要:目的:通过建立新生小鼠缺氧缺血性脑损伤(HIBD)模型和体外小鼠海马神经元系HT22细胞糖氧剥夺(OGD)模型,探讨淫羊藿苷(ICA)对新生小鼠脑组织以及HT22细胞卵泡抑素样蛋白1(FSTL1)表达的影响.方法:将36只7 d C57BL/6小鼠随机分为对照组(control)、HIBD组(HIBD)和HIBD+ICA组(HIBD+ICA),每组12只,采用改良的Rice-Vannucci法建立新生小鼠HIBD模型;体外将HT22细胞分为对照组(control)、OGD组、OGD+ICA组,建立HT22细胞OGD模型,采用免疫荧光染色分别检测FSTL1在新生小鼠HIBD脑组织以及HT22细胞内的表达情况,Western Blot检测ICA对新生小鼠HIBD脑组织以及HT22细胞FSTL1表达的影响.结果:体内体外实验结果均显示,新生小鼠遭受HIBD或HT22细胞遭受OGD损伤后,新生小鼠脑组织以及HT22细胞FSTL1表达水平显著升高,HT22细胞形态结构不完整,而给予ICA预处理后这一情况得到逆转,且HT22细胞形态得到明显恢复.结论:ICA可能通过促进FSTL1表达对新生儿HIBD发挥神经保护作用.Objective: By establishing hypoxic-ischemic brain damage(HIBD) model of neonatal mice, and an in vitro model of mouse hippocampal neuron line HT22 cell glucose and oxygen deprivation(OGD). To explore the effect of icariin(ICA) on the expression of follistatin-like 1(FSTL1) in the brain tissue of newborn mice and HT22 cells. Methods: In vivo, thirty-six 7 d C57 BL/6 mice were randomly divided into the control group(control), HIBD group, and HIBD+ICA group, with 12 mice in each group. A modified Rice-Vannucci method was used to establish a neonatal mouse HIBD model. In vitro, HT22 cells were divided into control group(control), OGD group, OGD+ICA group, and an OGD model of HT22 cells were established. Immunofluorescence staining was used to detect the expression of FSTL1 in neonatal mouse HIBD brain tissue and HT22 cells, and Western Blot was used to detect the effect of ICA on the expression of FSTL1 in neonatal mouse HIBD brain tissue and HT22 cells. Results: The results of in vivo and in vitro experiments showed that after newborn mice suffered from HIBD or HT22 suffered OGD damage, the expression level of FSTL1 in mouse brain tissue and HT22 cells was significantly increased, and the morphology and structure of HT22 cells were incomplete. After pretreatment with ICA, this situation was reversed, and the morphology of HT22 cells was significantly restored. Conclusion: ICA may play a neuroprotective effect on neonatal HIBD by promoting the expression of FSTL1.
