摘  要:大叶铁线莲具有较高的观赏价值和药用价值,利用SSR分子标记技术挑选出一套多态性好、稳定性强的SSR引物对合理开发和利用大叶铁线莲有着重要的意义。本试验筛选出了18对多态性较好的引物,并对这些引物的多态性进行分析。结果显示,18对引物在20个大叶铁线莲个体中一共获得了122个等位基因(Na),平均等位基因为6.8,观测杂合度(Ho)介于0.278~1.000之间,期望杂合度(He)介于0.465~0.917之间,多态位点信息含量(PIC)介于0.398~0.885之间,平均值为0.669。此外,利用NTSYS version2.10e软件绘制出大叶铁线莲个体的聚类分析树状图,结果表明,这18对SSR引物能够将基于不同居群的大叶铁线莲较好的分为两组,不同居群的大叶铁线莲之间也表现出了高度的遗传分化。本研究所开发的多态性SSR引物为大叶铁线莲以及铁线莲属植物的群体遗传学研究提供了研究工具。Clematis heracleifolia DC. has high ornamental value and medicinal value. It is of great significance to select a set of SSR primers with good polymorphism and strong stability by using SSR molecular marker technology for rational development and utilization of Clematis heracleifolia. In this experiment, 18 pairs of primers with better polymorphism were selected and the polymorphism of these primers was analyzed. The results showed that a total of122 alleles(Na) were obtained with 18 pairs of primers in 20 Clematis heracleifolia individuals, the average number of alleles was 6.8, the observed heterozygosity(Ho) was between 0.278~1.000, the expected heterozygosity(He) was between 0.465~0.917, the polymorphic locus information content(PIC) was between 0.398~0.885, and the average was 0.669. Using NTSYS version2.10 e software, the cluster analysis tree diagram of Clematis heracleifolia was drawn,and the results showed that these 18 pairs of SSR primers could better divide Clematis heracleifolia into two groups based on different populations, and there was also a high degree of genetic differentiation between Clematis heracleifolia based on different populations. The polymorphic SSR primers developed in this study can provide research tools for population genetic studies of Clematis heracleifolia and Clematis L.
