摘 要:目的:研究姜黄素不同时期给药对肠缺血再灌注损伤的保护作用及其作用机制。方法:将24只SD大鼠随机分为4组,假手术组、I/R模型组、姜黄素预处理组、姜黄素组,每组6只。肠系膜上动脉夹闭1 h,放开再灌注2 h造模。姜黄素预处理组:100 mg·kg^(-1)姜黄素小肠缺血前30 min腹腔注射;姜黄素组:100 mg·kg^(-1)姜黄素小肠缺血后30 min腹腔注射。取肠黏膜组织进行HE染色和Chiu评分评价病理学变化,ELISA法检测肿瘤坏死因子(TNF)-α、8-异前列腺素(PG)F2α、超氧化物歧化酶(SOD)、丙二醛(MDA)的表达水平。免疫印迹法检测Toll样受体(TLR)4和丝裂原活化蛋白激酶(MAPK)的蛋白表达水平。结果:与模型组比较,姜黄素预处理和姜黄素组肠组织病理学变化明显减轻,Chiu评分明显下降(P<0.05);姜黄素预处理和姜黄素组肠组织TNF-α、8-iso-PGF2α、MDA表达水平降低,SOD活性升高,TLR4、MAPK蛋白水平升高(P<0.05)。结论:姜黄素不同I/R时期给药处理均能发挥抗氧化和抗炎作用,均可改善大鼠肠组织缺血再灌注损伤。Objective:To investigate the protective effects of Curcumin against the intestinal ischemia-reperfusion injury(IRI)before and after IRI and its mechanism.Methods:24 SD rats were assigned to four groups in a random way:I/R model group,sham operation group,curcumin pretreatment group and curcumin group.I/R model group:after fasting for 24 h,SD rats were anesthetized by intraperitoneal injection of 10%chloral hydrate(3 mL/kg).Superior mesenteric artery(SMA)was separated and clipped along the midline of abdomen for 1 h,followed by reperfusion for 2 h.In sham operation group,only SMA separation was performed without plugging.Curcumin pretreatment group:100 mg·kg^(-1) curcumin was intraperitoneally injected 30 min before intestinal ischemia.Curcumin group:100 mg·kg^(-1) curcumin was intraperitoneally injected 30 min after intestinal ischemia.HE staining and Chiu’s scores were used to evaluate intestinal histological changes.TNF-α,8-iso-prostaglandin(PG)F2α,MDA contents and SOD activity of intestinal tissue were detected by ELISA.Protein expressions of Toll-like receptor(TLR)4 and mitogen-activated protein kinase(MAPK)were determined by Western blot.Results:Curcumin pretreatment and post-treatment could ameliorate IR-induced intestinal injury.Chiu’s scores in above two groups were remarkably decreased compared with model group(P<0.05).Levels of TNF-α,8-iso-PGF2α,and MDA in intestinal tissue were reduced and SOD activity was raised in Curcumin pretreatment and post-treatment groups.Protein levels of TLR4 and MAPK were remarkably elevated in model group(P<0.05)to compare with sham group.but decreased in Curcumin pretreatment and post-treatment groups(P<0.05).Conclusion:Curcumin can improve rat’s intestinal ischemia-reperfusion injury before and after IRI by anti-oxidative and anti-inflammatory effects.
