摘 要:[目的]针对广东玉米品种建立高效遗传再生技术体系。[方法]以穗甜1号、广糯1号和广黑甜3个玉米品种的幼胚为外植体,研究N_(6)培养基中不同浓度的L-Pro、AgNO_(3)、2,4-D对愈伤组织诱导率和胚性愈伤率的影响。[结果]较高浓度的Pro(1000~1300 mg/L)有利于玉米愈伤组织诱导,而继代时在N_(6)培养基中添加29.43μmol/L AgNO_(3)则可获得较多高质量的胚性愈伤组织;对3个玉米品种而言,培养基中2,4-D的适宜浓度为1~2 mg/L。3种基因型的外植体均可诱导出愈伤组织,但基因型和激素种类与浓度均影响愈伤组织诱导效率。[结论]在实际应用中,应根据不同基因型选择最佳的L-Pro、AgNO_(3)和2,4-D浓度配比,以获得较多高质量的胚性愈伤组织。[Objective]To establish high-frequency regeneration technique system for gene tranformation in maize in Guangdong.[Method]The effects of different concentrations of L-Pro,AgNO_(3) and 2,4-dichlorophenoxyacetic acid(2,4-D)in N_(6) medium on both the callus induction rate and embryogenic callus rate were analyzed with the immature embryos of 3 maize varieties Suitian 1,Guangnuo 1 and Guangheitian used as materials.[Result]The results showed that the percentage of inducing callus was increased evidently with the addition of L-Proline(1000-1300 mg/L),and it was optimal for improving the induction percentage of embryogenic callus to add AgNO_(3)(29.43μmol/L)in N_(6) medium during subculture;In addition,for 3 tested maize varieties,the most suitable concentration of 2,4-dichlorophenoxyacetic acid(2,4-D)was 1-2 mg/L callus were grown from immature embryos of all the three genetypes,but both genotypes and types and concentrations of phytohormone affcted the frequency of embryogenic callus formation.[Conclusion]In practice,the optimal concentrations of L-Pro,AgNO_(3) and 2,4-dichlorophenoxyacetic acid(2,4-D)were necessary to determined for different genotypes in callus induction in order to increase the induction percentage of embryogenic callus.
