新型鸭呼肠孤病毒山东株的分离鉴定及致病性分析

摘　　要：本试验采用病毒分离培养、RT-PCR扩增、测序分析、致病性试验等方法,以探索山东菏泽地区商品肉鸭养殖场内樱桃谷鸭脾脏坏死、瘸腿症状的病因。结果显示,鸡胚培养发病鸭的肝脏和脾脏组织悬液分离出1株病毒;该病毒株S 1基因与新型鸭呼肠孤病毒的同源性为94%~96%,与鹅呼肠孤病毒的同源性为95%,与番鸭呼肠孤病毒的同源性为49%~50%,与禽呼肠孤病毒同源性为44%~48%,从而确定该病毒株为新型鸭呼肠孤病毒;该病毒株经尿囊腔接种于7日龄SPF鸡胚后,胚体在72~96h内集中死亡,出现水肿、出血症状;该病毒株在Vero细胞上可产生明显的空泡化病变;该病毒株在1日龄雏鸭可回归出与临床一致的脾脏坏死,攻毒后第8周,可观察到50%的鸭发生瘸腿,同时肌肉和关节处可剖检到干酪样物。由此判断,新型鸭呼肠孤病毒感染是导致该养殖场鸭脾脏坏死和瘸腿的主要原因。This study utilized virus culture,RT-PCR,sequencing analysis,and virulence assay to establish the cause of spleen necrosis and lameness in Cherry Valley ducks,Shandong province.The results showed that chicken embryo culture of the liver and spleen extract from dead duck yielded a virus isolate;the S 1 gene of the virus isolate shared 94%-96%homology with that of novel duck reovirus strain,95%homology with that of goose orthoreovirus strain,49%-50%homology with that of Muscovy duck reovirus strain,44%-48%homology with that of avian orthoreovirus strain,confirming the virus isolate as novel duck reovirus strain.Inoculation of the virus isolate in 7-day-old SPF chicken embryo via the urethra cavity led to embryo death between 72 and 96 h,along with signs of edema and hemorrhage.Inoculation of the virus isolate onto Vero cells generated significant cavitation lesions.Inoculation of the virus isolate in 1-day-old duck caused spleen necrosis consistent with clinic cases,and 50%of ducks developed lameness in the 8th week,in addition to cheese-like substances in the muscles and joints.Thus,novel duck reovirus(NDRV)appeared to be the cause of spleen necrosis and lameness in Cherry Valley ducks,Shandong province.
