摘 要:目的研究黄芪在高糖诱导的血管平滑肌细胞(VSMCs)增殖和迁移中的作用及其机制。方法取VSMCs随机分为对照组(普通培养)、高糖组(25mmol·L^(-1)葡萄糖)、实验组(25 mmol·L^(-1)葡萄糖+40μg·m L^(-1)黄芪)、第1转染组(转染阴性对照寡核苷酸+25 mmol·L^(-1)葡萄糖+40μg·mL^(-1)黄芪)、第2转染组(转染miR-18a-5p模拟物+25 mmol·L^(-1)葡萄糖+40μg·m L^(-1)黄芪)。以细胞计数试剂盒8(CCK-8)法检测细胞的存活率;以划痕实验检测细胞迁移情况;以实时定量反转录聚合酶链反应(qRT-PCR)法检测miR-18a-5p的表达;以蛋白质印迹法检测线粒体融合蛋白2 (MFN2)蛋白的表达。结果对照组、高糖组和实验组VSMCs存活率分别为(100.00±10.00)%,(188.24±18.17)%和(123.52±12.66)%;VSMCs迁移率分别为(15.16±1.64)%,(53.63±5.27)%和(34.46±3.58)%;miR-18a-5p的表达分别为1.03±0.09,1.63±0.15和1.36±0.02;MFN2蛋白表达为1.05±0.12,0.34±0.07和0.73±0.09。以上数据,对照组和高糖组相比,高糖组和实验组相比,差异均有统计学意义(均P<0.05)。此外,第1转染组和第2转染组中MFN2蛋白的表达量为0.69±0.08和0.43±0.06,差异有统计学意义(P<0.05)。结论黄芪通过调控miR-18a-5p/MFN2轴抑制高糖诱导的VSMCs增殖和迁移。Objective To investigate the role of Astragalus in the proliferation and migration of vascular smooth muscle cells (VSMCs)induced by high glucose and its mechanism.Methods VSMCs were randomly divided into control group (normal culture),high glucose group(25 mmol·L^(-1)D-glucose),experimental group (25 mmol·L^(-1)D-glucose+40μg·mL^(-1)astragalus),transfection group 1 (transfection of negative control oligonucleotide+25 mmol·L^(-1)D-glucose+40μg·mL^(-1)astragalus),transfection group 2 (transfected with miR-18a-5p mimic+25 mmol·L^(-1)D-glucose+40μg·mL^(-1)astragalus).Cell counting kit-8 (CCK-8) assay was used to detect cell proliferation;scratch assay was used to detect cell migration;real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR)analysis was used to detect the expression of miR-18a-5p;Western blot was used to detect mitochondrial fusion protein 2 (MFN2) protein expression.Results The inhibition rates of VSMCs proliferation in control group,high glucose group and experimental group were (100.00±10.00)%,(188.24±18.17)%and (123.52±12.66)%,respectively;the migration rates of VSMCs were (15.16±1.64)%,(53.63±5.27)%and (34.46±3.58)%,respectively;the expressions of miR-18a-5p were 1.03±0.09,1.63±0.15 and 1.36±0.02,respectively;MFN2 protein expression were 1.05±0.12,0.34±0.07 and0.73±0.09.For the above data,there was statistically significant difference between control group and high glucose group,between high glucose group and experimental group (all P<0.05).In addition,the expression of MFN2protein in transfection group 1 and transfection group 2 groups were 0.69±0.08 and 0.43±0.06,and the difference was statistically significant (P<0.05).Conclusion Astragalus inhibits high-glucose induced proliferation and migration of VSMCs by regulating the miR-18a-5p/MFN2 axis.
