环状RNA CD151对过敏性鼻炎大鼠鼻黏膜上皮细胞炎症的影响

摘　　要：目的探究环状RNA CD151(circ CD151)通过调控微小RNA-30a-5p(miR-30a-5p)对过敏性鼻炎大鼠鼻黏膜上皮细胞炎症和黏液产生的影响。方法将SD大鼠随机分为对照组和模型组,其中对照组大鼠腹腔注射和滴鼻0.9%氯化钠溶液;模型组大鼠腹腔注射和鼻腔滴入卵清白蛋白构建过敏性鼻炎大鼠模型,并制备大鼠鼻黏膜上皮细胞,并将大鼠鼻黏膜上皮细胞分为6组:对照组,模型组,pc-NC组(转染pc-NC),pc-circ CD151组(转染pccirc CD151)、si-NC(转染si-NC)组和si-circ CD151(转染si-circ CD151)组。用酶联免疫吸附(ELISA)法检测粒细胞-巨噬细胞集落刺激因子(GM-CSF)、嗜酸性粒细胞趋化因子(Eotaxin)、干扰素-γ(IFN-γ)、白细胞介素12 (IL-12)以及黏蛋白5AC(MUC5AC)、黏蛋白5B(MUC5B)的含量;用实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测细胞中miR-30a-5p的表达;用双荧光素酶报告实验验证circ CD151与miR-30a-5p的结合关系。结果对照组,模型组,pc-NC组,pc-circ CD151组、si-NC组和si-circ CD151组中GM-CSF的含量分别为(115.78±2.56),(305.77±3.21),(303.12±3.47),(451.35±5.21),(302.09±2.88),(216.89±4.17) pg·m L^(-1);Eotaxin的含量分别为(211.45±2.71),(644.33±3.67),(647.35±3.71),(745.82±3.21),(639.38±3.91),(445.72±2.72) pg·m L^(-1);IFN-γ的含量分别为(101.24±5.66),(50.17±3.22),(48.21±3.36),(35.61±4.25),(53.63±3.26),(72.45±4.72) pg·m L^(-1);IL-12的含量分别为(61.11±3.47),(35.54±2.78),(33.47±2.55),(18.52±3.31),(37.15±2.56),(50.41±3.62) pg·m L^(-1);MUC5AC的含量分别为(205.35±15.78),(814.12±12.33),(816.72±11.56),(937.12±10.58),(809.18±8.99),(424.80±7.39) pg·m L^(-1);MUC5B的含量分别为(27.31±1.22),(65.47±3.29),(66.29±4.22),(85.39±3.67),(63.12±4.77),(47.29±3.88)μg·m L^(-1)。以上指标,对照组和模型组相比,pc-NC组和pc-circ CD151组相比,si-NC组和si-circ CD151组相比,差异均有统计学意义(均P<0.05)。circ CD151可负向调控miR-30a-5p的表达;双荧光素酶报告基�Objective To investigate the effect of circular RNA CD151(circCD151) on inflammation and mucus production in nasal mucosal epithelial cells of rats with allergic rhinitis through regulation of microRNA-30a-5p (miR-30a-5p).Methods SD rats were randomly divided into control group and model group,rats in control group were injected intraperitoneally and nasal drops of 0.9%NaCL^(-1)rats in model group were injected intraperitoneally and nasal drops of ovalbumin to construct allergic rhinitis rat model,and rat nasal mucosal epithelial cells were prepared.The nasal epithelial cells were divided into six groups:control group,model group,pc-NC group (transfected with pc-NC),pc-circCD151group (transfected with pc-circCD151),si-NC group (transfected with si-NC) and si-circ CD151 group(transfected with si-circCD151);The levels of granulocyte-macrophage colony-stimulating factor (GM-CSF),eosinophil chemokine (Eotaxin),interferon-gamma (IFN-γ),interleukin 12 (IL-12),and mucin 5AC(MUC5AC) and mucin 5B (MUC5B) were measured by enzyme-linked immunosorbent assay (ELISA).Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) experiment was used to detect the expression of miR-30a-5p in cells.A dual-luciferase reporter assay was used to verify the binding of circCD151 to miR-30a-5p.Results The GM-CSF levels in control,model,pc-NC,pc-circCD151,si-NC and si-circCD151 groups were(115.78±2.56),(305.77±3.21),(303.12±3.47),(451.35±5.21),(302.09±2.88),(216.89±4.17)pg·mL^(-1),the contents of Eotaxin were (211.45±2.71),(644.33±3.67),(647.35±3.71),(745.82±3.21),(639.38±3.91),(445.72±2.72) pg·m L^(-1),the contents of IFN-γwere (101.24±5.66),(50.17±3.22),(48.21±3.36),(35.61±4.25),(53.63±3.26),(72.45±4.72) pg·m L^(-1),the contents of IL-12 were(61.11±3.47),(35.54±2.78),(33.47±2.55),(18.52±3.31),(37.15±2.56),(50.41±3.62)pg·m L^(-1),the contents of MUC5AC were (205.35±15.78),(814.12±12.33),(816.72±11.56),(937.12±10.58),(809.18±8.99),(424.80±7.39) pg·mL^(-1),the contents of MUC5B were (27.31±1.
