摘  要:目的:探究链脲佐菌素(STZ)联合单侧肾切除手术诱导糖尿病肾病(DN)大鼠模型建立的方法,并对其有效性进行评价;观察当归补血汤对本研究中DN大鼠模型的肾组织保护作用。方法:采用灌胃脂肪乳及单次腹腔注射STZ(40 mg/kg),并结合单侧肾切除手术建立DN大鼠模型,观察建模期间受试动物的体质量及摄食量情况,分别在建模成功后的第4、6、8周检测各组受试动物的尿量、体质量,计算肾体比,计算UACR,检测相关血液生化学指标;造模成功的受试动物分为对照组、模型组、当归补血汤组(4.2 g/kg)及厄贝沙坦组(0.0017 g/kg),分别在给药第4、8周,计算肾体比,检测UACR、UREA、TG、CHOL、RBG等指标,并观察肾脏组织病理形态学改变。结果:DN模型制备过程中逐渐出现多饮、多食、多尿且体质量减轻,血糖升高的糖尿病典型症状,成模率为85%;在DN模型的第4、6、8周,与对照组比较,体质量、肾体比、摄食量、尿量、UACR及血液生化指标均有显著差异(P<0.01);当归补血汤的干预实验中,给药4、8周后,与DN模型组相比,DBD组的体质量明显升高(P<0.05)、肾体比明显降低(P<0.05,P<0.01),UACR显著降低(P<0.01),UREA、TG、CHOL、RBG等血液生化学指标明显下降(P<0.05),肾组织的病理形态学变化有明显改善。结论:SD大鼠灌胃给予脂肪乳,单次腹腔注射STZ并结合单侧肾切除手术,可成功制备DN动物模型,模型成功率较高,具有良好的稳定性及持续性,且受试动物的死亡率低;此动物模型通过当归补血汤的干预可降低RBG、调节TG、CHOL,改善肾功能,起到改善肾组织损伤的作用,从而有效控制糖尿病肾病的发展。Objective:To study the model establishment of diabetic nephropathy(DN)by streptozotocin(STZ)combined with unilateral nephrectomy in rats,and to evaluate the effectiveness of the model.To observe the protective effect of Danggui Buxue Decoction on renal tissue of DN rat model.Methods:The DN rat model was established by intragastric fat emulsion and single intraperitoneal injection of STZ(40 mg/kg)combined with unilateral nephriectomy.The weight and food intake of the experimental animals were observed during the modeling period.Urine volume and body weight,the renal body ratio and UACR,as well as blood biochemical indicators were calculated and detected at the 4th,6th and 8th week after the modeling respectively in each group.The successful models were divided into the model group,the Danggui Buxue Decoction group(4.2 g/kg)and the Irbesartan group(0.0017 g/kg),and the normal rats were taken as the blank control.At the 4th and 8th week of administration,the renal body ratio was calculated,the indicators of UACR,UREA,TG,CHOL and RBG were detected,and the renal histopathological changes were observed in the groups.Results:During the preparation of DN model,typical diabetic symptoms,such as polydipsia,polyphagia,polyuria,low body weight and elevated blood glucose,appeared gradually,and the successful modeling rate was 85%.At the 4th,6th and 8th week of DN modeling,there were significant differences in body weight,renal body ratio,food intake,urine volume,UACR and blood biochemical indexes between the model group and the control group(P<0.01).After 4 weeks and eight weeks of administration,the body weight was significantly increased(P<0.05),the renal body ratio was significantly decreased(P<0.05,P<0.01),the UACR was significantly decreased(P<0.01),the biochemical indexes of UREA,TG,CHOL and RBG were significantly reduced in the Danggui Buxue Decoction group compared with those in the model group(P<0.05);the pathological changes of renal tissues were significantly improved.Conclusion:SD rats were establised DN model by
