摘 要:中药多糖是由醛糖或酮糖通过糖苷键连接而成的天然高分子多聚物。由于其在生命过程中重要的生理功能及其广泛的应用被学者日益关注,引起了学者浓厚的兴趣,对其结构的研究是阐述其多种生物活性机制的前提。甲基化分析是确定糖苷键连接的最常用且有效的分析方法,该过程包括将多糖的单个组分衍生成部分甲基化阿尔迪醇乙酸酯衍生物,通过GC-MS分析和定性可以确定每个组成糖的连接方式。本文总结了甲基化分析在6种常见的中药多糖结构研究中的应用,从而阐述甲基化分析对其结构研究的重要作用,为甲基化分析在更多的中药多糖结构研究领域提供参考。Chinese medicinal polysaccharides are natural macromolecules composed of aldoses or ketoses linked by glycosidic linkages.Due to its important physiological functions in the life process and its wide application,it has been increasingly concerned and aroused strong interest by scholars.The study of its structure is the premise of elaborating its various biological activity mechanisms.Methylation analysis is the most common and efficient analytical method for determining glycosidic linkages,and the process involves derivatization of individual components of polysaccharides into partially methylated alditol acetate derivatives(PMAAs),through GC-MS and qualitative analysis,it is possible to determine how each constituent sugar is linked.This study summarized the application of methylation analysis in the structural research of six kinds of Chinese medicinal polysaccharides,thus expounding the important role of methylation analysis in its structural research,and providing a reference for methylation analysis in more structural research fields of Chinese medicinal polysaccharides.
