银翘解毒软胶囊体外抑制人冠状病毒229E及其介导的炎症的研究

摘　　要：目的探究银翘解毒软胶囊(YQ)对人冠状病毒229E(HCoV-229E)的体外抑制作用。方法通过经典的体外细胞模型分析银翘解毒软胶囊的体外毒性及对HCoV-229E的抑制作用;通过空斑实验验证银翘解毒软胶囊对HCoV-229E的抑制作用;应用实时定量PCR法分析银翘解毒软胶囊对HCoV-229E介异的炎症因子表达水平的影响。结果银翘解毒软胶囊对人肝癌细胞的50%毒性浓度为(3 315±131)μg·mL^(-1),对HCoV-229E的50%抑制浓度为(361.9±36)μg·mL^(-1),选择指数为9.16。空斑实验表明银翘解毒软胶囊2 mg·mL^(-1)几乎可完全抑制病毒复制(P<0.001)。银翘解毒软胶囊在2 mg·mL^(-1)浓度作用下,可明显抑制由冠状病毒HCoV-229E感染介导的炎症因子如趋化因子(C-C基元)配体5(CCL-5,P<0.001)、单核细胞趋化蛋白1(MCP-1,P<0.001)、白细胞介素6(IL-6,P<0.001)、白细胞介素8(IL-8,P<0.001)和肿瘤坏死因子α(TNF-α,P<0.001)核酸的升高。结论体外实验显示银翘解毒软胶囊可抑制人冠状病毒HCoV-229E的复制及其介导的炎症反应,具有作为抗冠状病毒药物的开发价值。Objective To explore in vitro inhibitory effect of Yinqiao Jiedu soft capsules on human coronavirus 229E(HCoV-229E).Method The toxicity of Yinqiao Jiedu soft capsules and its inhibitory effect on HCoV-229E in vitro were evaluated through the classic cell model.Plaque assay was performed to verify the inhibition of Yinqiao Jiedu soft capsules on HCoV-229E.Afterwards,real-time quantitative PCR was used to analyze the effect of Yinqiao Jiedu soft capsules on the expression of inflammatory factors induced by HCoV-229E.Results The 50%toxic concentration of Yinqiao Jiedu soft capsules on human liver cancer cells is(3315±131)μg·mL^(-1),the 50%inhibitory concentration on HCoV-229E is (361.9±36)μg·mL^(-1),and the selection index is 9.16.Plaque assay showed that Yinqiao Jiedu soft capsules(2 mg·mL^(-1))almost completely inhibited virus replication in vitro(P<0.001).Yinqiao Jiedu soft capsules at a concentration of 2 mg·mL^(-1)can significantly inhibit the increase of inflammatory factors induced by HCoV-229E infection,such as mRNA expressions of C-C motif chemokine ligand 5(CCL-5,P<0.001),monocyte chemotactic protein-1(MCP-1,P<0.001),interleukin-6(IL-6,P<0.001),interleukin-8(IL-8,P<0.001) and tumor necrosis factor-α(TNF-α,P<0.001).Conclusion In vitro Yinqiao Jiedu sof capsules can inhibit the replication of HCoV-229E and the inflammatory response induced by virus infection,which has development value as an anti-coronavirus drug.
