翠雀花素对急性心肌梗死小鼠心脏的保护作用

摘  要:目的研究翠雀花素对急性心肌梗死(AMI)小鼠心脏的保护作用。方法48只C57 BL/6J小鼠随机分为假手术组,模型组,翠雀花素低、高剂量组。采用永久性结扎冠状动脉左前降支构建小鼠AMI模型,术后翠雀花素低、高剂量组分别腹腔注射翠雀花素25、50 mg·kg^(-1),假手术组和模型组腹腔注射等容积溶剂。术后第7日采用小动物超声检测心脏功能,TTC染色检测心肌梗死面积,HE染色观察心肌病理改变,TUNEL染色检测心肌细胞凋亡水平,Western blot法检测心肌组织中凋亡和自噬相关蛋白的表达水平。结果与假手术组相比,模型组左室射血分数(LVEF)、左室短轴缩短率(LVFS)、左室收缩末期前壁厚度(LVAWs)和舒张末期前壁厚度(LVAWd)均显著降低(P<0.01),左室收缩末期内径(LVIDs)和舒张末期内径(LVIDd)显著增加(P<0.01),心肌梗死面积达(43.09±4.06)%,心肌细胞凋亡率显著增加(P<0.01),凋亡相关蛋白cleaved caspase 3表达增加、Bcl-2/Bax降低(P<0.01),自噬相关蛋白p62和LC3Ⅱ/LC3Ⅰ的表达无显著变化(P>0.05)。与模型组相比,翠雀花素低、高剂量组LVEF、LVFS、LVAWs和LVAWd均提高,LVIDs和LVIDd降低(P<0.01),心肌梗死面积显著降低(P<0.01),心肌病理损伤改善,心肌细胞凋亡率显著降低(P<0.01),cleaved caspase 3表达降低、Bcl-2/Bax增加(P<0.01),p62蛋白表达降低、LC3Ⅱ/LC3Ⅰ增加(P<0.01)。结论翠雀花素可改善AMI小鼠心脏功能,减少心肌梗死面积,减轻心肌病理损伤和细胞凋亡,其机制可能与增强心脏自噬有关。AIM To study the protective effect of delphinidin on the heart in mice with acute myocardial infarction(AMI)mice.METHODS Forty-eight C57 BL/6 J mice were randomly divided into sham group,model group,low and highdose delphinidin group.Permanent ligation of the left anterior descending coronary artery was used to construct AMI model.The low and high-dose delphinidin groups were intraperitoneally injected with delphinidin 25 and 50 mg·kg^(-1),respectively.The sham group and model group were intraperitoneally injected with equal volume of solvent.On the 7 th day after operation,cardiac function of mice was observed by ultrasonic echocardiography.TTC staining was used to detect the infarct size.HE staining was used to detect the damage of myocardial tissue.TUNEL staining was used to detect the level of apoptosis.Western blotting was used to detect the expression levels of apoptosis and autophagy-related proteins in myocardial tissues.RESULTS Compared with the sham group,the left ventricular ejection fraction(LVEF),short axis shortening rate(LVFS),systolic anterior wall thickness(LVAWs)and diastolic anterior wall thickness(LVAWd)reduced in the model group(P<0.01),while the left ventricular end-diastolic internal diameter(LVIDs)and end-diastolic volume(LVIDd)increased(P<0.01),the infarct size was(43.09±4.06)%,and the myocardial cell apoptosis rate increased significantly(P<0.01).The expression of apoptosis-related protein cleaved caspase 3 increased,Bcl-2/Bax decreased(P<0.01),and autophagyrelated protein p62 and LC3Ⅱ/LC3Ⅰexpression had no signifieant changes(P>0.05).Compared with the model group,LVEF,LVFS,LVAWs and LVAWd increased,and LVIDs and LVIDd decreased in the low and high-dose delphinidin groups(P<0.01).Meanwhile,the myocardial infarct size reduced(P<0.01),myocardial pathological damage improved,and myocardial cell apoptosis rate reduced significantly(P<0.01).The protein expression of cleaved caspase 3 and p62 decreased,while Bcl-2/Bax and LC3Ⅱ/LC3Ⅰincreased(P<0.01).CONCLUSION Delphinidin can improve
