摘 要:目的采用网络药理学方法及实验研究探讨复方刺梨合剂抗胃癌的作用机制。方法通过检索TCMSP数据库,获得复方刺梨合剂主要活性成分和相关作用靶点,通过CTD和OMIM数据库筛选抗胃癌靶点,利用Cytoscape 3.7.1软件构建药物-成分-靶点网络图,获取蛋白质相互作用关系,绘制蛋白关系网络,采用WebGestalt和DAVID数据库进行GO功能和KEGG通路分析,采用Western blot法测定p53、Bcl-2蛋白表达。结果共筛选了复方刺梨合剂10个活性化学成分和236个化合物作用靶点,得到440个胃癌相关靶点和38个靶标,GO功能涉及对刺激反应、生物调节、细胞传递等,KEGG通路分析涉及63条显著信号通路,可见复方刺梨合剂通过参与调控多种生物学过程而发挥抗癌作用,实验结果证实复方刺梨合剂使用后p53和Bcl-2蛋白表达增加。结论基于网络药理学方法及实验研究,揭示了复方刺梨合剂抗胃癌作用是多成分、多靶点、多通路作用的结果,可能通过调控p53和Bcl-2蛋白的表达发挥作用。Objective To explore the mechanism of the compound Cili mixture anti-gastric cancer activity using the network pharmacology analysis and then validate some of the mechanisms in vitro.Methods The traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)was used to identify the main active components and related targets of the compound Cili mixture,while their anti-gastric cancer targets were obtained from the Comparative Toxicogenomics Database(CTD)and Online Mendelian Inheritance in Man(OMIM)databases.Next,the software of Cytoscape 3.7.1 was used to construct its network diagram of the compound,for the component and targets and protein-protein interaction network.The Gestalt and David databases were used to analyze the gene ontology(GO)terms and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway.After that,level of p53 and Bcl-2 expression was confirmed using Western blot.Results There were 10 main active chemical components in the compound Cili mixture and 236 compound targets identified for a total of 440 gastric cancer-related targets and 38 direct targets.The GO terms were in the response to stimulation,biological regulation,and cell communication,while the KEGG pathways were involved in 63 significant signaling pathways.The in vitro experiments showed that treatment of gastric cancer cells with the compound Cili mixture induced p53 and Bcl-2 levels in vitro.Conclusion The anti-gastric cancer activity of the compound Cili mixture was involved in multi-components,multi-targets and multi-gene signaling,indicating that the compound Cili mixture played an anti-cancer role by participating in the regulation of various biological processes.
