摘 要:目的:研究急性热应激对睾丸免疫微环境相关蛋白的影响。方法:49只雄性SD大鼠随机分为对照组和热应激组(0.5 h组、2 h组、4 h组、6 h组、24 h组和48 h组)。热应激组大鼠麻醉后43℃恒温水浴20 min,并分别于0.5 h、2 h、4 h、6 h、24 h和48 h后摘取睾丸。Western blot检测睾丸内occludin、ZO-1、vimentin、p-NF-κB p65和p-IκBα的表达;ELISA法检测睾丸内IL-1β表达。结果:与对照组相比,热应激组vimentin和occludin表达均显著降低(P<0.01);0.5 h组ZO-1表达差异无统计学意义(P>0.05),其余热应激组ZO-1表达显著降低(P<0.01);0.5 h组、2 h组和4 h组睾丸内p-NF-κB p65和p-IκBα表达与对照组相比差异无统计学意义(P>0.05),6 h组、24 h组和48 h组p-NF-κB p65和p-IκBα表达显著升高(P<0.01)。与对照组相比,24 h组和48 h组睾丸内IL-1β表达差异无统计学意义(P>0.05)。结论:急性热应激0.5 h后,细胞骨架结构和血睾屏障紧密连接结构即出现异常,且在48 h内损伤逐渐加重。睾丸内炎症相关NF-κB信号通路激活则发生在6 h以后,但并未导致睾丸内炎症因子增加,表明急性热应激可破坏睾丸免疫豁免结构,激活NF-κB信号通路,却不能诱发局部炎症反应。Objective:To study the effect of acute heat stress on immune microenvironment-related proteins in testis.Methods:Forty-nine adult male SD rats were randomly divided into control group and heat stress groups(0.5 h group,2 h group,4 h group,6 h group,24 h group and 48 h group).After heat stress group rats were bathed in a water bath at 43℃for 20 min,then testes were removed at 0.5 h,2 h,4 h,6 h,24 h and 48 h.Western blot was used to detect expressions of occludin,ZO-1,vimentin,p-NF-κB p65 and p-IκBαin testis;ELISA method was used to detect expression of IL-1βin testis.Results:Compared with control group,expressions of vimentin and occludin in heat stress groups were significantly decreased(P<0.01);expression of ZO-1 in 0.5 h group was not statistically significant(P>0.05),and ZO-1 in the other heat shock groups were significantly reduced(P<0.01);expressions of p-NF-κB p65 and p-IκBαin 0.5 h,2 h and 4 h groups were not significantly different(P>0.05);expressions of p-NF-κB p65 and p-IκBαin 6 h group,24 h group and 48 h group were significantly increased(P<0.01).Compared with control group,there was no statistically significant change in expressions of IL-1βof 24 h and 48 h groups(P>0.05).Conclusion:In 0.5 h after acute heat stress,the cytoskeleton structure and tight junction structure of blood testis barrier appear abnormal,and the damage gradually worsen within48 h.Activation of inflammation-related NF-κB signaling pathway in the testis occurred after 6 h.However,there is no increase of inflammatory factors in testis.Our data indicate that acute heat stress can destroy the immune immunity structure of testis,activate NF-κB signaling pathway,but does not cause inflammation in testis.
