摘  要:目的:探讨IL-4和IL-4R基因单核苷酸多态性和复发性流产(RSA)的关系。方法:选取2019年1月至2020年8月甘肃省妇幼保健院收治的150例RSA患者(RSA组)和同期150例女性健康志愿者(对照组),采用SNaPshot SNP分型技术检测外周血中IL-4 rs2243250和IL-4R rs1805010基因多态性,研究组间基因型分布差异,分析IL-4 rs2243250和IL-4R rs1805010基因多态性和RSA易感的相关性。结果:IL-4R rs1805010位点GA基因型和等位基因A分布在RSA和对照组间的差异有统计学意义(P<0.05)。携带有A等位基因的人群RSA发病风险降低(OR=0.71,95%CI=0.51~0.99),携带有AA和GA基因型的人群RSA发病风险低于GG基因型(OR=0.49,95%CI=0.29~0.85)。结论:IL-4R rs1805010位点A等位基因和AA、GA基因型可能是RSA发病的保护因素,但仍有必要进一步开展质量更高、样本量更大的病例对照研究进行验证。Objective:To investigate the association between single nucleotide polymorphisms of IL-4 and IL-4R genes and recurrent spontaneous abortion(RSA).Methods:150 RSA patients(RSA group)and 150 female healthy volunteers(control group)were selected from January 2019 to August 2020. IL-4 rs2243250 and IL-4R rs1805010 gene polymorphisms in peripheral blood were detected by SNaPshot SNP typing technique. Genotypic distribution differences among study groups were studied,and association between IL-4 rs2243250 and IL-4R rs1805010 gene polymorphisms and RSA susceptibility was analyzed.Results:There were significant differences in distribution of GA genotype and allele A at IL-4R rs1805010 locus between RSA and control groups(P<0.05). Risk of RSA was lower in people with A allele(OR=0.71,95%CI=0.51~0.99). Risk of RSA in people with AA and GA genotypes were lower than that of GG genotypes(OR=0.49,95%CI=0.29~0.85).Conclusion:A allele and AA and GA genotypes of IL-4R rs1805010 may be protective factors for RSA,but further case-control studies with higher quality and larger sample size are still necessary to verify.
