摘　　要：为了建立和优化马卵巢石蜡切片制作体系,为完善马属动物繁殖理论知识奠定基础,试验以2~3岁蒙古马为研究对象,选择6匹健康母马卵巢组织,对比了不同固定方法、组织脱水方法、浸蜡方法及苏木精-伊红(Hematoxylin and Eosin,H.E.)染色体系对石蜡切片制作效果的影响。结果表明:大于0.5 cm卵巢组织片段、经过3种固定液均达不到有效支持后期石蜡切片制作的条件;马卵巢石蜡切片制作需组织片段小于0.5 cm并采用Bouin固定液进行75%酒精浸泡24 h→85%酒精浸泡1 h→95%浸泡酒精2 h→100%酒精2 h脱水;二甲苯透明剂中放置10 min×2次;纯蜡与二甲苯按1∶1比例混合并浸蜡40 min→纯蜡Ⅰ40 min→纯蜡Ⅱ0.05 Mpa真空浸蜡40 min条件,以此为制作马卵巢组织石蜡切片的最佳体系。In order to establish and optimize the paraffin section production system of the horse ovary,and to further improve the knowledge of equine reproduction theory,the experiment took 2 to 3-year-old Mongolian horses as the research object,selected 6 healthy mares ovarian tissues,and the effect of dehydration method,immersion wax method,and hematoxylin and eosin( Hematoxylin and Eosin,H. E.) staining system on the production of paraffin sections were detected and compared. The results showed that: ovarian tissue fragments larger than 0. 5 cm and after 3 kinds of fixatives could not meet the conditions to effectively support the later stage of paraffin section preparation;The preparation of equine ovarian paraffin section requires tissue fragments less than 0. 5 cm and 75% alcohol immersion in Bouin fixative for 24 h →85% alcohol immersion for 1 h→95% alcohol immersion for 2 h→100% alcohol for 2 h for dehydration;placed in xylene transparent agent for 10 min × twice;pure wax and xylene were mixed in a ratio of 1∶1 and immersed in wax for 40 min →Pure wax Ⅰ 40 min→Pure wax Ⅱ 0. 05 Mpa vacuum immersion wax for 40 min is the best system for making paraffin sections of horse ovarian tissue.
