HPLC测定藿香正气制剂中2种抗病毒类成分的含量

摘　　要：目的建立高效液相色谱法(HPLC)测定藿香正气系列制剂中2种抗病毒类成分槲皮素和芹菜素的含量。方法色谱条件采用Luna-C_(18)(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.4%磷酸溶液为流动相进行梯度洗脱,流速1.0 ml/min,检测波长360 nm,柱温30℃。结果槲皮素、芹菜素的线性范围分别为0.643~128.6μg/ml(r=0.9999),0.511~102.2μg/ml(r=0.9997);平均加样回收率分别为98.68%和100.34%;RSD分别为1.72%和1.93%。不同制剂中2种成分含量相差较大,藿香正气水中槲皮素含量最高,藿香正气丸剂中芹菜素含量最高,藿香正气合剂中未检测到槲皮素和芹菜素。结论本试验所建立的方法准确可靠、重复性好,可为藿香正气制剂的质量控制提供科学的依据。Objective To establish an HPLC method for simultaneous determination of quercetin and apigenin in Huoxiang Zhengqi serial preparations.Methods Two flavonids were separated on Luna-C_(18)(4.6 mm×250 mm,5μm)chromatography column using methanol-0.4%phosphoric acid as the flow phase for gradient elution.The flow rate was 1.0 ml/min,the detection wavelength was set at 360 nm,and the column temperature was 30℃.Results The linear ranges of quercetin and apigenin were 0.643-128.6μg/ml(r=0.9999),0.511-102.2μg/ml(r=0.9997),and the average recoveries were 98.68%and 100.34%,and the RSDs were 1.72%,1.93%,respectively.The content of quercetin in Huoxiang Zhengqi water was the highest,the content of apigenin in Huoxiang Zhengqi pill was the highest,and quercetin and apigenin were not detected in Huoxiang Zhengqi mixture.Conclusion The established method is accurate,reliable and reproducible,which can provide a scientific basis for the quality control of Huoxiang Zhengqi preparation.
