摘  要:为了探讨邻苯二甲酸二(2-乙基)己酯(DEHP)对斑马鱼早期生命阶段下丘脑-垂体-性腺轴(HPG)和下丘脑-垂体-肾上腺轴(HPA)相关功能基因表达的影响,将斑马鱼的受精卵暴露于10、30和300μg/L DEHP溶液中,同时设置空白对照,96 h后,利用实时荧光定量PCR技术,测定斑马鱼仔鱼HPG和HPA轴上12个功能基因的表达。结果显示,与对照组相比,30μg/L DEHP处理组的Cyp19b、GnRH3、Era和ERβ基因表达量显著升高,GnRH2、Pomc、Mr、Mc2r、Crh、Gr和Crhbp基因在各DEHP处理组表达量显著降低,FSH-β和Crhbp基因表达量无显著变化。以上研究结果表明:DEHP对斑马鱼胚胎/仔鱼阶段的2个主要内分泌分子通路产生了显著影响,将会导致内分泌功能的改变,从而可能影响斑马鱼的后期生长发育。Danio rerio fertilized eggs were exposed to the blank control and three different concentrations(10μg/L,30μg/L,and 300μg/L)of di(2-ethylhexyl)phthalate(DEHP)for a 96 h exposure to investigate the effects of DEHP on functional gene expression of hypothalamus-pituitary-gonadal(HPG)and hypothalamus-pituitary-adrenal(HPA)axes of D.rerio during the early life stage.Then the mRNA expression levels of functional genes related to HPG and HPA axes were determined by real-time fluorescence quantitative PCR(RT-qPCR)techniques.The results showed that compared with the control group,the mRNA expression levels of Cyp19b,Gnrh3,Er-a,and ER-βgenes were significantly increased in 30μg/L DEHP treated group.In contrast,the mRNA levels of GnRH2,Pomc,Mr,Mc2r,Crh,and Gr genes were significantly decreased in all DEHP treated groups.FSH-βand Crhbp genes mRNA levels were not significantly changed.The results showed that the changes of two major endocrine molecular pathways induced by DEHP exposure in embryo/larvae stages of D.rerio could cause changes in endocrine function,which may affect the growth and development of D.rerio at later stage.
