摘 要:为了研究姜黄提取散中姜黄素在草鱼体内的药代动力学及组织分布规律,对草鱼(Ctenopharyngodon idellus)按每千克鱼体重单剂量口灌100 mg姜黄提取散后,采用液相色谱-质谱联用法测定不同时间点草鱼各组织中姜黄素含量,用药代动力学分析软件(WinNonlin5.2.1)处理药-时数据。结果显示:该检测方法在0.05~50.00μg/L范围内线性关系良好,相关系数为0.9991,血浆中检测限为0.02μg/L,其它组织中为0.02μg/kg,方法平均回收率为77.16%~106.10%,日间、日内误差均小于10%。姜黄素在血浆中达峰时间(T_(max))为8 h,峰浓度(C_(max))为0.406μg/L,药时曲线下面积AUC为5.947 h·μg/L,消除半衰期(t_(1/2))为21.596 h,姜黄素在组织中分布呈现出组织特异性。结果表明,姜黄素在草鱼血液中达峰时间长,峰浓度低,消除快,在体内主要分布于肝脏和肾脏。The concentration of curcumin in each tissue was determined by liquid chromatography-mass spectrometry to study pharmacokinetics and tissue distribution of curcumin of Jiang huang ti qu San in Ctenopharyngodon idellus.The data were processed by WinNonlin5.2.1 pharmacokinetic software after oral administration with a single dose of 100 mg/kg body weight.The results showed that the liquid chromatography-mass spectrometry has a linear correlation in the concentration range of 0.05~50.00μg/L,with a correlation coefficient of 0.9991.The quantitative limits of thiamphenicol were 0.02μg/L in plasma and 0.02μg/kg in muscle,skin,kidney,and liver,respectively.The average recovery was 77.16%~106.10%,and the intra-and inter-day precisions were lower than 10%.The T_(max) was 8.00 h,the C_(max) was 0.406μg/L,the AUC was 5.947 h·μg/L,the t_(1/2) was 21.596 h in plasma.And distribution of curcumin in grass carp exhibited apparent tissue specificity.After oral administration,it indicated long peak time,low peak concentration,and rapid elimination in grass carp blood.And the curcumin concentration was found to be highest in liver and kidney.
