精液中冷冻保护剂加入比例对冷冻复苏后精子活动力的影响研究

摘　　要：目的 探讨精液中冷冻保护剂加入比例对冷冻复苏后精子活动力的影响。方法 20例人类精子库精子捐献志愿者,以手淫法留取精液4 ml,按照1︰3分为甲组(1 ml)、乙组(3 ml)。制备蛋黄甘油复合型冷冻保护剂并依据1︰1、1︰3的保护剂/精浆配比向两组样本中各加入1 ml冷冻保护剂。对比两组冷冻前、冷冻复苏后前向运动精子百分比、精子活力。结果 冷冻前,两组前向运动精子百分比对比差异无统计学意义(P>0.05);冷冻复苏后,两组前向运动精子百分比均较冷冻前明显下降,但乙组前向运动精子百分比(42.36±4.63)%高于甲组的(33.48±4.56)%,差异具有统计学意义(P<0.05)。冷冻前,两组精子活力对比差异无统计学意义(P>0.05);冷冻复苏后,两组精子活力均较本组冷冻前明显下降,但乙组精子活力(55.64±3.12)%高于甲组的(52.14±3.08)%,差异具有统计学意义(P<0.05)。结论 精液中冷冻保护剂加入比例影响冷冻复苏后精子活动力,相比于保护剂/精浆配比1︰1,配比1︰3复苏后精子活动力保护的更好,下降幅度更小,减少了冷冻带来的精子活动力损伤。Objective To discuss the effect of cryoprotectant proportion in semen on sperm motility after cryo-resuscitation.Methods 20 volunteers were selected from the human sperm bank,and 4 ml semen was collected by masturbation method and divided into group A(1 ml)and group B(3 ml)according to the ratio of 1︰3.The egg yolk glycerin complex cryoprotectant was prepared,and 1 ml cryoprotectant was added to each of the two groups according to the ratio of 1︰1 and 1︰3 cryoprotectant/seminal plasma.The percentage of forward motile sperm before freezing and after cryoresuscitation,and sperm motility were compared between the two groups.Results Before freezing,there was no statistically significant difference in the percentage of forward motile sperm between the two groups(P>0.05).After cryoresuscitation,the percentage of forward motile sperm in the two groups were significantly decreased compared with those before freezing;the percentage of forward motile sperm(42.36±4.63)%in group B were significantly higher than(33.48±4.56)%in group A;all the differences were statistically significant(P<0.05).Before freezing,there was no statistically significant difference in sperm motility between the two groups(P>0.05).After cryoresuscitation,the sperm motility of the two groups were obviously decreased compared with those before freezing;the sperm motility(55.64±3.12)%in group B were significantly higher than(52.14±3.08)%in group A;all the differences were statistically significant(P<0.05).Conclusion The proportion of cryoprotectant in semen affects the sperm motility after cryoresuscitation.Compared with the ratio of cryoprotectant/seminal plasma 1︰1,the ratio of 1︰3 has better protection of sperm motility after resuscitation,and the decline level is lower,which reduces the damage of sperm motility caused by cryopreservation.
