摘　　要：目的:建立高效液相色谱法同时测定北部湾海鸭蛋蛋黄中虾青素、叶黄素、玉米黄素、角黄素和β-胡萝卜素5种天然类胡萝卜素含量,同时对比罗氏比色扇法测定结果,合理分析出天然类胡萝卜素总含量与蛋黄色泽两者之间的关联。方法:样品经皂化后用复合溶剂[石油醚+乙醚=1+1(V/V)]提取,用水洗涤去碱性,旋蒸干后用乙腈复溶,Athena C30 HPLC Column(250 mm×4.6 mm,5μm)色谱柱分离,以甲醇+1%磷酸水+叔丁基甲醚作为流动相进行梯度洗脱,流速1.0 mL·min^(-1),DAD二极管阵列检测器,检测波长474 nm。结果:5种类胡萝卜素可以良好分离,相关系数r2均在0.997以上,样品在3个浓度水平的加标回收率为86.7%~98.4%(n=8),精密度为1.59%~3.78%。结论:该方法快速实用,测定的色素总量结果与采用罗氏比色扇法测定色度结果具有明显的关联,色素总量越高,其色度值也越高,可以作为海鸭蛋中天然色素的有效检测方法,克服了罗氏法易受个人主观性影响的缺陷,具有更科学、更准确的优点。Objective:This paper establishes a high performance liquid chromatography method to simultaneously determine the content of five natural carotenoids in Beibu Gulf sea duck egg yolk,astaxanthin,lutein,zeaxanthin,canthaxanthin andβ-carotene,and compare the results of the Roche colorimetric fan method and reasonably analyze the relationship between the total content of natural carotenoids and the yolk color of the egg.Method:After saponification,the samples were extracted with composite solvent[petroleum ether+ether=1+1(V/V)],washed with water to remove alkalinity,evaporated and redissolved with acetonitrile,Athena C30 HPLC Column(250 mm×4.6 mm,5μm)chromatographic column separation,gradient elution with methanol+1%phosphoric acid water+tert butyl methyl ether as mobile phase,flow rate 1.0 mL·min^(-1),DAD diode array detector,detection wavelength 474 nm.Result:The five carotenes could be well separated,and the linear correlation coefficient r^(2) was more than 0.997.The recovery of spiked samples at three concentration levels was 86.7%~98.4%(n=8),and the precision was 1.59%~3.78%.Conclusion:This method is fast and practical,the total amount of pigment determined has an obvious correlation with the chromaticity determined by Roche colorimetric fan method,the higher the total amount of pigment,the higher the chromaticity value.It can be used as an effective method for the detection of natural pigment in sea duck eggs,overcomes the defect that Roche method is easy to be affected by personal subjectivity,and has the advantages of more science and accuracy.
