摘  要:目的研究SARS-CoV-2病人恢复期的病毒特异性的记忆性T细胞和抗体水平的变化。方法通过SARS-CoV-2特异性抗原肽刺激COVID-19病人来源PBMC,流式细胞术检测不同疾病严重程度COVID-19患者SARS-CoV-2特异性的记忆性CD8^(+)T细胞的数目以及功能,ELISA检测不同疾病严重程度COVID-19患者血清中S1、S2特异性的IgG水平。结果与中、重症COVID-19患者相比,无症状或轻症的COVID-19患者体内拥有数目更多,质量更好的SARS-CoV-2特异性的记忆性CD8^(+)T细胞;中、重症SARS-CoV-2感染患者血清中病毒特异性IgG水平随着时间延长而降低,维持时间较短。结论无症状和轻症COVID-19患者体内拥有质量更好的病毒特异性的记忆性CD8^(+)T细胞。The ongoing pandemic of COVID-19,caused by SARS-CoV-2 infection,has caused a worldwide emergency.Thus far,there is no study about the changes of SARS-CoV-2-specific memory CD8^(+)T cells and IgG level in China COVID-19 patients.This study was performed to study the changes of SARS-CoV-2 specific memory CD8^(+)T cells and IgG level in COVID-19 patients during convalescence.Peripheral blood mononuclear cell(PBMC)was isolated and stimulated by SARS-CoV2-specific CD8^(+)T cells peptide,then the number and function were detected by flow cytometry.Furthermore,S1 and S2 specific IgG levels in serum of COVID-19 patients were detected by ELISA.Data showed that the quantity and quality of SARS-COV-2-specific memory CD8^(+)T cells in asymptomatic or mild COVID-19 patients were higher than those in moderate or severe COVID-19 patients.The level of virus-specific IgG in serum of patients with moderate and severe SARS-COV-2 infection decreased over time,only maintained for a short time.All above data suggested that asymptomatic or mild COVID-19 patients have a good quality of virus-specific memory CD8^(+)T cells.
