摘 要:为了探究Tc1/Mariner转座子超家族对褶皱臂尾轮虫(Brachionus plicatilis)基因组结构和基因表达调控的影响,本研究基于褶皱臂尾轮虫的基因组和转录组数据,对褶皱臂尾轮虫的重复序列进行了注释,并对褶皱臂尾轮虫的转座子的表达和其临近基因的表达进行了统计和富集,在基因上下游及内部区域富集到了10个转座子家族(其中3个家族属于Tc1/Mariner超家族)。在针对褶皱臂尾轮虫的Tc1/Mariner超家族分析中,共发现其7个亚家族:Tc1(DD34E)、Tc2(DD35D)、Mariner(DD34D)、Pogo(DDxD)、Sagan(DD30D)、Tigger(DD32/36D)和Fot1(DD30D),并主要分布在基因间区,对转座酶催化区域的结构和系统发育分析显示拥有完整转座酶的3个亚家族(Tc1、Tc2和Pogo)保守性强且聚类情况良好。基于转录组数据发现各个家族的表达模式较为相似,在雄性阶段表达较高。Tc1/Mariner超家族临近基因的功能大量涉及到环境信息处理和生物发育调节,反映了褶皱臂尾轮虫基因组对环境的适应。本研究通过对褶皱臂尾轮虫Tc1/Mariner转座子超家族在基因组中的进化和表达进行系统分析,为从功能角度认知Tc1/Mariner转座子并理解Tc1/Mariner对基因组进化的作用提供了新的线索。To explore the role of Tc1/Mariner in the genome construction and process of gene expression regulation of Brachionus plicatilis,the genome and transcriptome data was used to mine and enrich the expression of the transposon of B.plicatilis genome and analyze the expression of adjacent genes.Ten transposon families within and near the genes were identified,of which 3 belong to Tc1/Mariner superfamily.Analysis of the Tc1/Mariner superfamily revealed a total of 7 subfamilies: Tc1(DD34E),Tc2(DD35D),Mariner(DD34D),Pogo(DDxD),Sagan(DD30D),Tigger(DD32/36D)and Fot1(DD30D),which mainly distributed in the intergenic region.The structural and phylogenetic analysis of the catalytic region of transposase showed that three subfamilies(Tc1,Tc2 and Pogo)possessing complete transposases were highly conserved and clustered well.The transcriptome data showed that the expression patterns of all families were relatively similar,and the expression was higher in the male stage.The functions of adjacent genes in the Tc1/Mariner superfamily involved environmental information processing and biological development regulation,reflecting the adaptive evolution of the B.plicatilis genome.This systematic study concluded a comprehensive understanding for the distribution and expression of Tc1/Mariner transposon as well as their adjacent genes,which will provide novel insight into the role of Tc1/Mariner on the evolution of the B.plicatilis genome.
