摘 要:为全面了解白桦类甜蛋白(thaumatin-like protein, TLP)基因家族成员的结构、功能和表达特性,本研究采用同源序列比对法,从白桦基因组数据库中筛选并鉴定到41个类甜蛋白基因家族成员,分别命名为BpTLP1~BpTLP41,对其进行生物信息学分析,并通过半定量对白桦TLP家族成员的组织表达特异性进行了分析。结果表明,41个白桦类甜蛋白家族成员分别定位在11条染色体上,其中8号染色体分布最多,包括14个家族成员。该家族成员的氨基酸数量在336~975 aa之间,分子量在28 054.11~81 966.03 kD之间,理论等电点在5.01~5.21之间,偏酸性。基因半定量结果表明,41个白桦TLP家族成员在根、茎、叶中均有不同程度的表达,其中,BpTLP35、BpTLP39在叶片中表达量较高,Bp TLP14、BpTLP35、BpTLP36在茎中表达量较高,BpTLP41、BpTLP3在根中表达量较高。In order to comprehensively understand the structure, function and expression characteristics of thaumatin-like protein(TLP) gene family members in B. platyphylla, 41 TLP gene family members were screened and identified from B. platyphylla genome database by homologous sequence alignment method, and they were named as BpTLP1~BpTLP41, respectively. The bioinformatics analysis was carried out, and the tissue expression specificity of TLP family members in B. platyphylla was analyzed by semi-quantitative analysis. The results showed that the sugarbeet protein family members were distributed on 11 chromosomes, of which chromosome 8 was the most distributed, with 14. The number of amino acids ranged from 336 to 975 aa, the molecular weight ranged from28 054.11 to 81 966.03 kD, and the theoretical isoelectric point ranged from 5.01 to 5.21. So it is acidic amino acid. Forty-one B. platyphylla TLPs were expressed in roots, stems and leaves to varying degrees. Among them,BpTLP35 and BpTLP39 were highly expressed in leaves, BpTLP14, BpTLP35 and BpTLP36 were highly expressed in stems, and BpTLP41 and BpTLP3 were highly expressed in roots.
