摘 要:为探究羊草种子休眠特性,对赤霉素处理后的羊草种子进行转录组测序,共检测到了37 208条Unigene,序列总长为40 055 874 bp,并使用MISA软件对所有的Unigene进行SSR搜索,共检测到3 771个SSR位点,每10.6 kb就会出现一个SSR位点。重复基元类型从单核苷酸至六核苷酸类型均有出现,重复基元以三核苷酸为主,占比56.91%,其次为二核苷酸,占比24.71%。在155种重复基元中,重复基元数量以CCG/CGG类型最多,占比为18.38%,其次为AG/CT,占比14.35%。SSR重复次数主要集中在4~8次,数量为3 076个,占比81.57%。SSR长度范围为12~259 bp,主要集中在12~24 bp,其中SSR长度≥20 bp的高多态性SSR数量为1 017个,占比26.97%。本研究有助于羊草的遗传多样性及分子育种研究。In order to explore the dormancy characteristics of Leymus chinensis seeds, transcriptome sequencing was performed on Leymus chinensis seeds treated with GA3. A total of 37 208 Unigenes with a total sequence length of 40 055 874 bp were detected. MISA software was used to search all Unigenes by SSR, and a total of3 771 SSR sites were detected, one SSR site appears every 10.6 kb. The types of repetitive motifs range from mononucleotide to hexanucleotide. The repetitive motifs are mainly trinucleotides, accounting for 56.91%,followed by dinucleotides, accounting for 24.71%. Among the 155 repetitive primitives, CCG/CGG has the largest number of repeated primitives, accounting for 18.38%, followed by AG/CT, accounting for 14.35%. The number of SSR repetitions is mainly concentrated in 4~8 times, the number is 3 076, accounting for 81.57%. The length of SSR ranged from 12 to 259 bp, mainly concentrated in 12 to 24 bp. Among them, the number of highly polymorphic SSRs with SSR length ≥20 bp was 1 017, accounting for 26.97%. This research is helpful for genetic diversity and molecular breeding research of Leymus chinensis.
