摘 要:为了掌握拉萨地区羊多杀性巴氏杆菌(Pasteurella multocida,Pm)的感染情况和主要流行基因型,试验采用细菌分离培养、生化试验、PCR鉴定、药敏试验等方法对2018—2020年采自于拉萨地区的36份临床病料样品进行了Pm的分离鉴定,同时分析了其荚膜和脂多糖基因型。结果表明:纯化菌落经革兰氏染色可见革兰氏阴性菌,呈短杆状,共分离培养出8株菌;生化试验符合Pm的生化特性;PCR鉴定得到大小约为460 bp的目的条带;8株分离菌对氟苯尼考、复方新诺明、恩诺沙星、环丙沙星的敏感率分别为87.5%、75.0%、75.0%、62.5%,对卡那霉素和林可霉素的耐药率分别为87.5%和75.0%;8株分离菌中有4株为A:L3型,2株为B:L2型,1株为A:L6型,1株为D:L6型,共4种基因型。说明拉萨地区Pm的流行基因型存在一定程度的多样性,以A:L3为主要流行基因型,同时存在B:L2型、A:L6型、D:L6型的流行。In order to grasp the infection status and main epidemic genotypes of Pasteurella multocida(Pm) in sheep in Lhasa area, the experiment used the methods of bacterial isolation and culture, biochemical test, PCR identification, and drug susceptibility test to isolate and identify Pm from 36 clinical diseased material samples collected in Lhasa area from 2018 to 2020. Its capsule and lipopolysaccharide genotypes were also analyzed. The results showed that Gram-negative bacteria could be seen in the purified colony after Gram staining, which was in the shape of short rods and a total of eight strains were isolated and cultured. The biochemical test accorded with the biochemical characteristics of Pm;PCR identified a target band with a size of about 460 bp. The sensitivity rates of the 8 isolated strains to florfenicol, compound trimethoprim, enrofloxacin, and ciprofloxacin were 87.5%, 75.0%, 75.0%, and 62.5%, respectively, and the drug resistance rates to kanamycin and lincomycin were 87.5% and 75.0%, respectively. Among the 8 strains, 4 strains were A: L3;2 strains were B: L2;1 strain was A: L6;1 strain was D: L6, with a total of 4 genotypes. The results suggested that there was a certain degree of diversity in the epidemic genotypes of Pm in Lhasa area, with A: L3 as the main epidemic genotype, and meanwhile there were epidemics of B: L2, A: L6, and D: L6.
