血清SGK1基因多态性与冠心病患者疾病严重程度的相关性及预后分析

摘  要:目的:探讨与分析血清/糖皮质激素调节激酶(serum/glucocorticoid regulated kinases,SGK)1基因多态性与冠心病患者疾病严重程度的相关性及预后分析。方法:选择2018年8月-2021年1月在大连市第三人民医院诊治的冠心病患者78例作为冠心病组,选择同期健康体检人群78例作为对照组,检测两组血清SGK1含量与SGK1基因表达水平,同时进行血清SGK1基因多态性分析,调查冠心病患者的疾病严重程度、预后并进行相关性分析。结果:冠心病组血清SGK1含量与SGK1基因相对表达水平均低于对照组(P<0.05)。冠心病组SGK1基因 rs2267018位点GG基因型频率高于对照组(P<0.05),等位基因G频率高于对照组(P<0.05)。在冠心病组中,平均Gensini评分为(46.28±6.11)分。随访到2021年3月,出现主要心血管不良事件21例,发生率为26.9%。相关分析显示,Gensini评分与SGK1基因rs2267018位点GG基因型、等位基因G、血清SGK1含量、SGK1基因表达水平均呈正相关(P<0.05)。结论:冠心病患者多伴随血清SGK1表达水平降低,多由于SGK1基因 rs2267018位点基因多态性造成,GG基因型、等位基因G是影响患者疾病严重程度的重要危险因素。Objective:To investigate and analyze the correlation between serum/glucocorticoid regulated kinases (SGK)1 polymorphism and disease severity and prognosis in patients with coronary heart disease.Method:From August 2018 to January 2021,78 cases of patients with coronary heart disease who were diagnosed and treated in the Third People’s Hospital of Dalian were selected as the coronary heart disease group.At the same time,78 cases of healthy patients were selected as the control group,and the serum SGK1 content and SGK1 gene expression level of the two groups were detected.At the same time,the serum SGK1 gene polymorphism were analyzed,the disease severity and prognosis were investigated,and correlation was analyzed.Result:The serum SGK1 content and the relative expression level of SGK1 gene in the coronary heart disease group were lower than those in the control group (P<0.05).The frequency of GG genotype at rs2267018 locus of SGK1 gene in coronary heart disease group was higher than that of control group (P<0.05),and the frequency of allele G was higher than that of control group (P<0.05).In the coronary heart disease group,the average Gensini score were (46.28±6.11) points.Followed-up until March 2021,there were 21 cases of major adverse cardiovascular events occurred,with the incidence rate of 26.9%.Correlation analysis showed that Gensini score were positively correlated with SGK1 gene rs2267018 locus GG genotype,allele G,serum SGK1 content and SGK1 gene expression level (P<0.05).Conclusion:Patients with coronary heart disease are mostly accompanied by decreased serum SGK1 expression levels,which are mostly caused by the polymorphism of SGK1 gene rs2267018 locus,GG genotype and allele G are important risk factors that affect the severity of the disease and prognosis.
