高效液相色谱法测定胃炎颗粒中的去甲异波尔定和橙皮苷

摘　　要：目的建立同时测定胃炎颗粒中两种指标性成分的方法,用于胃炎颗粒成品的质量控制和评价;方法使用高效液相色谱法,对胃炎颗粒成品中去甲异波尔定和橙皮苷进行同时测定;色谱柱选择Waters XBridge C_(18)(250×4.6 mm,5μm),流动相为乙腈(A)-0.2%甲酸,梯度洗脱,程序为:0~15 min,10%~25%A;15~30 min,25%~95%A;30~35 min,95%~10%A;流速1.0 mL·min^(-1),检测波长280nm,进样量10μL,柱温25℃,共检测10批供试品;结果10批样品中,橙皮苷为0.8272~1.0965 mg·g^(-1),去甲异波尔定的含量为0.3031~0.5925 mg·g^(-1);结论方法快速、准确,能够对胃炎颗粒进行质量和工艺评价。OBJECTIVE To establish a method for simultaneous determination of two kinds of index components in WeiYan granules for quality control and evaluation of finished WeiYan granules.METHODS Normethopordine and hesperidin in WeiYan granules were determined simultaneously by HPLC,using Waters XBridge C_(18) column(250×4.6 mm,5μm)with mobile phase consisted of acetonitrile(A)-0.2%formic acid and gradient elution;the gradient program was 0-15 min,10%~25%A;15-30 min,25%~95%A;30-35 min,95%~10%A;The flow rate was 1.0 mL·min^(-1),the detection wavelength was 280nm,the injection volume was 10μL,and the column temperature was 25℃.A total of 10 batches of samples were detected.RESULTS In 10 batches of samples,the contents of hesperidin and normetopolidine were 0.8272-1.0965 mg·g^(-1) and 0.3031-0.5925 mg·g^(-1) respectively.CONCLUSION The method is rapid,accurate and can evaluate the quality and process of WeiYan granules.
