摘 要:目的检测右美托咪定(dexmedetomidine,DEX)对小鼠听性稳态反应(auditory steady-state response,ASSR)的影响,以探讨DEX对大脑感觉中枢神经电活动的调节作用。方法选用8只6~8周龄C57BL/6小鼠,全身麻醉下,在小鼠听皮层(auditory cortex,AC)区进行慢性电极植入手术,恢复1周后,使用在体电生理记录方法,分别记录每只小鼠在自由清醒状态下和腹腔注DEX(80μg/kg)后,对40 Hz click-trains声音刺激的ASSR变化;用平均能量(mean trial power,MTP)和锁相因子(phase-locking factor,PLF)对ASSR进行定量评估。结果腹腔注射DEX后可以显著降低40Hz ASSR的MTP和PLF(t7=2.858,P<0.05;t7=2.507,P<0.05)。结论DEX诱导的镇静效应可能与大脑皮层神经元之间同步活动减少相关,而ASSR可用于监测DEX镇静效果的潜在生物标志物。Objective To monitor the effect of dexmedetomidine(DEX)on the cortical the auditory steady-state response(ASSR)using a translational animal model and reveal the regulation of DEX on the electrical activity of the sensory central nervous system of the brain.Methods Under general anesthesia,chronic electrodes were implanted into the skull over the auditory cortex(AC)of male C57BL/6 mice(8 mice,6~8 weeks old).Animals recovered for 1 weeks after surgery,electrophysiological recording was conducted.Stimuli were 40 Hz click-trains presented when the mice were under free-moving condition.EEGs were recorded in AC of the same mouse before and after injection of DEX(80μg/kg)to monitor the changes of ASSR.Mean trial power(MTP)and phase-locking factor(PLF)were used to quantitatively evaluate the effect of DEX on ASSR.Results Data recorded from the AC of free moving mice showed that the MTP and PLF of 40 Hz ASSR were significantly reduced by the application of DEX(t7=2.858,P<0.05;t7=2.507,P<0.05,paired t-test).Conclusion Our results suggest that reduction of synchronized activity among the cortical neurons may be associated with the DEX-induced sedation,and ASSR is a potential biomarker for monitoring the sedative effect of DEX.
