摘 要:目的:建立西洋参细胞悬浮培养体系。方法:通过不同激素和物理影响因素筛选出西洋参细胞培养的最佳条件。结果:将西洋参愈伤组织接种于附加0.5mg·L^(-1)2,4-D+0.1 mg·L^(-1)KT+0.2 mg·L^(-1)NAA的MS培养基上最适合西洋参愈伤组织的生长;用西洋参愈伤组织制作西洋参细胞,发现西洋参悬浮细胞生长曲线类似“S”型;通过改变西洋参细胞悬浮培养的外界条件发现,西洋参悬浮细胞生长的最佳接种量为1.5 g/30mL,摇床转速和光照条件对西洋参细胞生长影响没有显著差异。结论:建立了西洋参细胞悬浮培养体系,该技术为西洋参细胞的扩大培养和有效成分的提取提供了物质基础。Establishing a suspension culture system of American ginseng cells.Methods:The best conditions of cell culture of Panax quinquefolium were screened out by different hormones and physical factors Results:The callus of Panax quinquefolium was inoculated with 0.5 mg·L^(-1)2,4-D+0.1 mg·L^(-1)KT+0.2 mg·L^(-1)NAA was the most suitable medium for the growth of Panax quinquefolium callus;using Panax quinquefolium callus to make Panax quinquefolium suspension cells,it was found that the growth curve of Panax quinquefolium suspension cells was similar to"s"type;by changing the external conditions of Panax quinquefolium cell suspension culture,it was found that the best inoculation amount for the growth of Panax quinquefolium suspension cells was 1.5 g/30mL.The growth of Panax quinquefolium cells was not statistically significant by light and the rotation speed of shaking table.Conclusion:The suspension culture system of Panax quinquefolium cells was established,which provided material basis for the expansion of cell culture and the extraction of effective components.
