摘  要:目的建立决明子总蒽醌(含游离蒽醌)的含量测定方法并采用正交试验法优化决明子的炮制工艺。方法采用差示分光光度法建立决明子总蒽醌(含游离蒽醌)的含量测定方法;以游离蒽醌含量与总蒽醌含量比值为综合评价指标,考察《中华人民共和国药典》2020年版收载的清炒、砂炒、蛤粉炒对决明子游离蒽醌和总蒽醌的含量及综合评价指标的影响。结果决明子总蒽醌(含游离蒽醌)的含量测定线性方程为△A=0.0501c-0.0015,r=0.9999,方法学验证符合要求;最佳的炒制方法为蛤粉炒,蛤粉炒工艺为武火、炒制时间为3min。结论差示分光光度法可用于决明子总蒽醌(含游离蒽醌)的含量测定,蛤粉炒过程受热均匀,决明子无煳化、焦化现象,工艺稳定,可用于决明子的炮制。Objective To establish a method for the determination of the content of total anthraquinone(including free anthraquinone)in Cassia seeds and to optimize the technology of processing Cassia seeds by orthogonal test.Methods The method for determining the content of total anthraquinone(including free anthraquinone)of Cassia seeds was established by differential spectrophotometry.Taking the ratio of free anthraquinone content to total anthraquinone content as the comprehensive evaluation indicator,this study investigated the effect of the methods(including pure stir-frying,sand stir-frying,and stir-frying with clam powder)collected in Pharmacopeia of the People’s Republic of China(2020 edition)on the contents of free and total anthraquinone and comprehensive evaluation indicator of Cassia seeds.Results The linear equation for determining the content of the total anthraquinone(including free anthraquinone)of Cassia seeds was△A=0.0501c-0.0015,r=0.9999.The methodology was verified to meet the requirements.The best stir-frying method was sand stir-frying with medium heat for 3 minutes.Conclusion Differential spectrophotometry can be used for the determination of the content of total anthraquinone(including free anthraquinone)in Cassia seeds.The sand stir-frying process is heated evenly and during the process the Cassia seed is free from burning and coking.Therefore,the technology is stable and can be used for processing Cassia seeds.
