摘  要:目的探讨荧光定量PCR检测结核杆菌DNA的拷贝数与涂片抗酸染色镜检阳性程度的相关性。方法选取可疑结核病患者1 012例,取TB-DNA标本1 012份,同时应用荧光定量PCR检测和涂片抗酸染色镜检,根据图片抗酸染色镜检结果进行分类,统计分析荧光定量PCR检测结核杆菌DNA的拷贝数。结果 1 012份标本中包括阳性标本241份,阴性标本771份;771份阴性标本中189份荧光定量PCR检测结果为阳性,结核杆菌DNA拷贝数大多为102~103数量级;241份阳性标本中223份荧光定量PCR检测阳性,结核杆菌DNA拷贝数大多为103~104数量级,与涂片阳性程度表现为正相关。结论荧光定量PCR检测结核杆菌DNA的拷贝数与涂片抗酸染色镜检阳性程度存在较为明显的相关性,可作为结核杆菌检测的有效方法。Objective To investigate the correlation between the copy number of Mycobacterium tuberculosis DNA detected by fluorescence quantitative PCR(FQ-PCR) and the positive degree of acid fast staining microscopy.Methods 1 012 TB-DNA samples were collected from 1 012 patients with suspected tuberculosis.Fluorescence quantitative PCR and acid fast staining microscopy were used to detect TB-DNA.According to the results of acid fast staining microscopy,the copies of TB-DNA detected by fluorescence quantitative PCR were analyzed statistically.Results Among 1 012 samples,241 were positive and 771 were negative;189 of 771 negative samples were positive by FQ-PCR,and most of the copies of Mycobacterium tuberculosis DNA were 102 to103.Among the 241 positive samples,223 were positive by FQ-PCR.The DNA copy number of Mycobacterium tuberculosis was mostly 103 to 104,which was positively correlated with the positive degree of smear.Conclusion There is a significant correlation between the copy number of Mycobacterium tuberculosis DNA detected by FQPCR and the positive degree of smear acid fast staining microscopy,which can be used as an effective method for the detection of Mycobacterium tuberculosis.
