摘 要:了解3种淫羊藿内生细菌群落的结构和组成,为淫羊藿内生菌资源的开发及利用提供参考。采用Illumina Miseq高通量测序技术,分析了粗毛淫羊藿、箭叶淫羊藿及巫山淫羊藿新叶和老叶内生细菌群落的结构和组成。3种淫羊藿叶片内生细菌群落组成结果显示,门水平的优势菌群主要包括变形菌门、厚壁菌门和放线菌门,优势菌属主要为甲基杆菌属(Methylobacterium)、鞘氨醇单孢菌属(Sphingomonas)、放线孢菌属(Actinomycetospora)、Kroppenstedtia属、泛菌属(Pantoea)及乳杆菌属(Lactobacillus)等。各样品的优势菌属(相对丰度>5%)存在一定差异,如在粗毛淫羊藿样品中甲基杆菌属、鞘氨醇单孢菌属相对丰度较其他样品高(相对丰度不足2%),而Kroppenstedtia属在箭叶淫羊藿样品中的相对丰度较高,乳杆菌属、魏斯式菌属(Weissella)、马赛菌属(Massilia)和泛菌属的相对丰度则在巫山淫羊藿样品中较高;同时,3种淫羊藿新叶和老叶内生细菌群落组成也存在差异,甲基杆菌属、鞘氨醇单孢菌属、Kroppenstedtia属和马赛菌属在老叶中的相对丰度较高,而乳杆菌属、魏斯式菌属和泛菌属在新叶中相对丰度较高,这些结果达到高度一致。优势益生菌的功能主要体现在促进土壤有机物的分解、合成多种次级代谢产物、诱导作物对病原微生物的抗性等方面。不同种淫羊藿叶片内生细菌群落组成不同,同种淫羊藿的新叶和老叶的内生细菌群落结构也存在差异。该研究首次分析了淫羊藿内生细菌的群落结构,为进一步研究淫羊藿内生细菌微生态及功能菌株的开发利用提供参考。Bacterial community composition and diversity in leaves of three kinds of Epimedium was revealed,aiming at providing scientific bases for the utilization of endophytic bacteria resources.High-throughput sequencing technology of Illumins Miseg was used to analyze the endophytic bacterial communities in new and old leaves of E.acuminatum,E.sagittatum,and E.wushanense.The results of community composition of Epeimedium showed that the dominant bacteria at phylum level mainly included Proteobacteria,Firmicutes,and Actinobacteria,and the dominant bacteria at genus level mainly Methylobacterium,Sphingomonas,Actinomycetospora,Kroppenstedtia,Pantoea,and Lactobacillus etc.There are certain differences in the dominant genus of each sample(relative abundance less than 5%),such as Methylobacterium and Sphingomonas in E.acuminatum belongs to the relatively high abundance than the rest of the samples(relative abundance of less than 2%),while Kroppenstedtia in E.sagittatum is higher,Lactobacillus,Weissella and Pantoea are higher in E.wushanense;At the same time,the endophytic bacterial community composition of new and old leaves of three Epimedium was also different.The relative abundance of Methylobacterium,Sphingomonas,Kroppenstedtia and Massilia in old leaves was higher,while the abundance of Lactobacillus,Weissella and Pantoea in new leaves was higher.These results were highly consistent.The functions of dominant endophytic bacteria communities reflected promoting the decomposition of soil organic matter,synthesizing a variety of secondary metabolites,and inducing crop resistance to pathogenic microorganisms and so on.The endophytic bacterial community composition of leaves of different kinds of Epimedium was different,and the new and old leaves of the same Epimedium was also different.This study was for the first time to analyze the community structure of endophytic bacteria in Epimedium,which provided scientific bases for the development and utilization of endophytic bacteria microecology and functional strains in Epimedii
