摘 要:目的:采用网络药理学分析大果木姜子主要挥发性成分治疗冠心病的分子机制。方法:通过顶空固相微萃取技术分析贵州大果木姜子挥发性成分,统计相对含量大于5%挥发性成分进行靶点筛选,通过Swiss Target Prediction对主要挥发性成分进行靶点筛选,在GeneCards数据库中以“Coronary heart disease”为关键词检索冠心病疾病靶点,基于R中clusterProfiler包进行GO(Gene ontology)和KEGG(Kyoto encyclopedia of genes and genomes)富集分析,通过STRING数据库检索靶点可能的潜在相互作用。结果:桧烯(15.54%)、桉树油(6.50%)、β-石竹烯(10.04%)相对含量大于5%,共筛选出126个靶点,基于GeneCards筛选出冠心病靶点1709个,通过FunRich取交集,筛选出大果木姜子治疗冠心病靶点66个,主要通过PPAR信号通路(PPAR signaling pathway)防治冠心病。通过蛋白相互作用分析发现alpha-2A肾上腺素能受体(Alpha-2A adrenergic receptor,ADRA2A)、alpha-2B肾上腺素能受体(Alpha-2B adrenergic receptor,ADRA2B)、Alpha-2C肾上腺素能受体(Alpha-2C adrenergic receptor,ADRA2C)、过氧化物异构酶体增殖子激活受体(Peroxisome proliferator-activated receptor gamma,PPAR-γ)为关键靶点。结论:大果木姜子主要挥发性成分防治冠心病的关键靶点为ADRA2A、ADRA2B、ADRA2C、PPAR-γ。Objective Using network pharmacology to analyze the molecular mechanism of the main volatile components of Litsea lancilimba Merr.in the treatment of coronary heart disease.Methods Analysis of volatile components of Litsea lancilimba Merr.by headspace solid phase microextraction technology,the relative content of volatile components were more than 5%,and the main volatile components were selected by Swiss Target Prediction.Search for coronary heart disease target with“Coronary heart disease”as key word in GeneCards database.Based on the clusterProfiler package in R for GO(Gene ontology)and KEGG(Kyoto encyclopedia of genes and genomes)enrichment analysis.Retrieving possible potential interactions of targets through STRING databases.Results The relative contents of sabinene(15.54%),eucalyptus oil(6.50%)andβ-bamboos(10.04%)were more than 5%.A total of 126 targets were screened.1709 coronary heart disease targets selected based on GeneCards.66 targets for the treatment of coronary heart disease in Litsea lancilimba Merr.were selected by FunRich.Prevention and treatment of coronary heart disease mainly through PPAR signaling pathway.Protein interaction analysis revealed alpha-2A adrenergic receptor(ADRA2A),alpha-2B adrenergic receptor(ADRA2B),Alpha-2C adrenergic receptor(ADRA2C)and Peroxisome proliferator-activated receptor gamma(PPAR-γ)are key targets.Conclusion The key targets of the main volatile constituents of the major volatile components in the prevention and treatment of coronary heart disease are ADRA2A,ADRA2B,ADRA2C and PPAR-γ.
