摘 要:目的:采用高效液相色谱(HPLC)法建立香莲祛痛霜血竭素含量的测定方法。方法:按照HPLC法(中国药典2015年版四部通则)测定。色谱条件与系统适用性试验色谱条件。色谱柱:AgilentC 18(250 mm×4.6 mm,5μm);流动相:乙腈-0.05 mol/L磷酸二氢钠溶液(45∶55);检测波长:440 nm;流速:1 mL/min;柱温:40℃;进样体积:10μL。结果:血竭素对照品溶液,以浓度对峰面积进行回归,在5.817~58.17μg/mL之间呈线形关系,平均峰面积675.2,RSD为0.35%;香莲祛痛霜平均峰面积314.7,RSD为1.36%;稳定性试验血竭素高氯酸盐对照品溶液均峰面积674.6,RSD为0.25%,样品中血竭素高氯酸盐平均峰面积362.2,RSD为2.32%;血竭素高氯酸盐的平均回收率为97.8%,RSD(n=9)为2.29%;三组不同批次的样品1 g血竭素含量为0.03248%。结论:本方法简便易行,结果准确,重复性好,可作为香莲祛痛霜质量控制标准使用。Objective To establish a High-Performance Liquid Chromatography(HPLC)method for the determination of dracodrin in Xiang Lian Qu Tong cream.Methods According to the four general principles of Chinese Pharmacopoeia 2015 edition.Chromatographic Conditions and System Applicability Test Chromatographic Conditions.Chromatographic Column:Agilent C 18(250 mm×4.6 mm,5μm);Mobile Phase:Acetonitrile-0.05 mol/L Sodium Dihydrogen Phosphate Solution(45∶55);Detection Wavelength:440 nm;Flow Rate:1 mL/min;Column;Temperature:40℃;Injection:10μL.Results dracorhodin reference solution showed a linear relationship between 5.817 and 58.17μg/mL,with an average peak area of 675.2 and RSD of 0.35%.The average peak area of Xianglian Qutong Cream was 314.7 and RSD was 1.36%.The average peak area of dracorhodin perchlorate reference solution was 674.6 and RSD was 0.25%.The average peak area of dracorhodin perchlorate was 362.2,RSD was 2.32%.The average recovery rate of dracorhodin perchlorate was 97.8%,RSD(n=9)was 2.29%.The content of dracorhodin in 1 g of three groups of samples was 0.03248%.Conclusion The method is simple,accurate and reproducible.It can be used as the quality control standard of Xianglian Qutong Cream.
