摘  要:内毒素耐受是指机体或细胞提前经低剂量脂多糖连续刺激后,对大剂量脂多糖的再刺激呈低反应甚至是无反应的一种现象。内毒素耐受是一个复杂的病理生理过程,涉及多个细胞信号通路、生物分子和受体改变。到目前为止,确切的机制尚不十分清楚。最近的研究表明内毒素耐受现象与一些疾病有关,如脓毒症、糖尿病、囊性纤维化和癌症等。本文就内毒素耐受的体内和体外模型、Toll样受体家族及其下游信号转导途径、表观遗传学及其对内毒素耐受机制的调控及临床应用研究进展做一综述。Endotoxin tolerance refers to the phenomenon that the body or cells show low response or even no response to the restimulation of high-dose lipopolysaccharide after continuous stimulation of low dose of lipopolysaccharide in advance.Endotoxin tolerance is a complex pathophysiological process involving multiple cellular signal pathways,biological molecules and receptor alternations.As yet,the exact mechanism remains unclear.Recent studies have shown that endotoxin tolerance is also associated with certain diseases such as sepsis,diabetes mellitus,cystic fibrosis,and cancer.This article reviews in vivo and in vitro models of endotoxin tolerance,toll-like receptor family and its downstream signal transduction pathways,epigenetics and its regulation of endotoxin tolerance mechanism,clinical application and research progress.
